Alfalfa industry as an important economic growth point in the southern mountain area of Ningxia is of great significance for the development of animal husbandry and the early realization of Ningxia's poverty shake off poverty.Alfalfa brown spot is one of the key factors affecting the healthy development of alfalfa industry in China.It caused by Pseudomonas medicaginis.The study of prevention and control of this disease is benefit to the development of alfalfa industry.Take P.medicaginis sporogenesis,nonsporogenesis fruiting body,and the leaves with P.medicaginis infection as the materials.The transcriptomics of P.medicaginis sporogenous and nonsporogenous fruiting body were studied,the relationship between them and proteomics was analyzed.The ultrastructures of P.medicaginis infected alfalfa leaves were observed.The effects of P.medicaginis infection on photosynthetic physiology of alfalfa leaves were analyzed.The results are as follows:(1)According to the transcriptome study of P.medicaginis sporogenesis and nonsporogenesis,776 differentially expressed genes mainly focus on protein processing,energy and transport metabolism.The analysis of the pathway of differential gene enrichment showed that the up-regulated genes were mainly involved in energy supply,the down-regulated genes were mainly involved in the synthesis of sugars.The 6 genes related to peptidylprolyl isomerase and DNA repair protein played important role in the sporulation of P.medicaginis.(2)The results of correlation analysis between transcriptomics and proteomics of P.medicaginis sporogenesis and nonsporogenesis showed that the proteins associated with genes were participitated in protein processing,carbon metabolism,ribosome and amino acid synthesis,oxidative phosphorylation and antibiotic biosynthesis.The multifunctional ?-oxidation protein,serine/threonine protein kinase,microtubule related protein,hypothetical hydrolase,two unknown protein and two hypothetical proteins showed similar trend at the transcription and protein levels.These genes played important role in the sporulation of P.medicaginis.(3)The results of ultrastucture observation showed that the mature fruiting body was black,breaking through the epidermal cells of the host,the ascorbic disc was dehiscent,and the ascorbic cap was absent.(4)The infected leaves by P.medicaginis showed Fv/Fm,?PS? and the photochemical quenching coefficient decreased significantly,at the same time NPQ increased.P.medicaginis limited photosynthetic related electronics supply by reduced the original photochemical efficiency of blade photosystem ? and electron transfer ability from antenna pigment to photosystem ?.P.medicaginis affected the growth of alfalfa by affecting the photosynthesis.
