| Objective: By establishing the model of the whole layer skin burns of dogs,the efficacy of bone marrow interstitial stem cell secretion factor to treat the whole layer of skin burns in dogs is discussed,and it is used to provide a basis for its further clinical application.Method: 1.12 healthy Chinese garden dogs were randomly divided into 3 groups with 4 dogs in each group.Group A: the dogs were burned with solid gasoline.Group B: experimental dogs were scalded with water at 95℃.Group C: dogs were treated with alcohol burns.After modeling,the number of deaths was recorded,the appearance of the burned part of the test dog was observed,and the whole skin was sampled to make pathological sections.According to the death rate,appearance,subcutaneous appearance after scab cutting and histopathological observation of the three groups after model building,the advantages and disadvantages of the three burn model building methods in all-layer skin model building were compared and analyzed.Determine the most suitable molding method for dogs with full skin burns.2.32 healthy dogs were selected and randomly divided into 4 groups(n=8),which were divided into the single modeling group(group A).Silver sulfadiazine treatment group(group B);Bone marrow mesenchymal stem cell secretory factor treatment group(group C);Blank control group(group D).The burn site was observed and recorded daily in each group after treatment.At 0,1,2,3,7,14,21 days after treatment,the contents of c-reactive protein and TNF-were determined by ELISA.Healing area and pathology were measured on days 0,7,14 and 21 after treatment.The obtained data were statistically analyzed using SPSS22.0 software.Result: 1.The mortality rate was 25% in group A,0% in group B and 0% in group C during modeling.Apparent indicators: observation group A and group B after building building skin uneven heating area,mild form to appear,gray dot focal necrosis(Ⅱ deep degree burns),swelling and other parts of the yellowish-white serous effusion(Ⅰdegree burns).Serious part is black dry scab(Ⅲ degree burns).Group B for 10 seconds when the light is light yellow to dark yellow skin redness,less resilient skin lesions(shallow Ⅱ degree burns).The burned area is serious when skin elasticity disappear entirely,lesions characterized by consistent dry necrosis,skin area of significant narrowing,outer crust covered(Ⅱ deep degree burns).Pathological indicators: in group A and group C,the skin structure was edema and denatured in each layer after 5-second burn,and the skin pathological sections were denatured and necrotic in each layer after 10-second,15-second and 20-second burn,with A large number of inflammatory cells infiltrating(full-layer skin burn).After 5 seconds of burns in group B,the whole epidermis was coagulated and necrotic or the epidermis was exfoliated,the dermis was edema,and the dermal structure of the skin was not clearly arranged after 10 seconds.After 15 and 20 seconds of modeling,the whole layer of structure in group B was blurred and there was a lot of inflammatory cell infiltration(full layer burn).Subcutaneous manifestations during scab cutting: in group A and group C,muscle redness and bleeding were found in the skin of the burned skin after 10,15 and 20 seconds of modeling.Muscle damage occurred at 20 seconds in group B.Comparison of results: the heating area of group A and group C could not be controlled,resulting in the unstable skin burn area and uneven burn degree after modeling,and vice versa for group B.To sum up,it can be concluded that to make a full-layer skin burn model of dog,the best result is to burn the model in 95℃ water for 15 seconds.2.The mortality rate during treatment was 37.5% in group A,12.5% in group B,no death in group C,and no death in group D.Changes in the appearance of the wound: on the 7th day after treatment,severe suppuration occurred in the single model group(group A).Sulfadiazine silver treatment group(group B)the wound surface was red and swollen,the yellow exudate congealed into scab.Bone marrow mesenchymal stem cell secretory factor treatment group(group C)reduced residual area,granulation tissue filling margin can be seen.After 14 days of treatment,the wound shrinkage of group A was more serious.In group B,the wound surface was dry and the scab was removed.After the scab was removed from the wound surface of group C,the wound healing was complete,and the skin tissue after healing was red and white.After 21 days of treatment,the wound surface of group A was not epithelial.The wound healing rate of group B was 91%,and that of group C was 100%.The healing period of group A(34.7000±6.53241)d,group B(29.2000±1.92304)d and group C(19.7300±2.23402)d was significantly lower than that of group A and group B(P<0.01).At the beginning of the second day of treatment,the level of TNF-in group C continued to decrease,the level of TNF-in group C was significantly lower than that in group A and group B(P<0.01),and the level of TNF-in group C was close to that in group D.C-reactive protein(CRP)was measured from the first day of treatment in three groups of experimental dogs,and there was no difference in CRP level among the three groups.On the second day of treatment,CRP levels in groups A,B and C began to decrease,and the difference between group C and group A and B was very significant(p < 0.01).On the 7th day,the CRP level of group C was close to that of group D.Pathological observation showed that capillaries in group C were abundant,a small amount of collagen fibers were close to the wound,and the wound surface was stratified from top to bottom.In group B,fewer capillaries were observed,inflammatory cell infiltration was obvious,and a small amount of fibroblast hyperplasia was observed.In group A,tissue necrosis was severe,skin stratification was not obvious and there was no epithelialization trend.After 14 days,the wound in group C was completely converged and epithelialized,with immature secretory gland tissue.In group B,the wound tissue was epithelialized and the blood vessels were not widely distributed.A large number of component fiber cells appeared,and epithelialization appeared in wound repair.After 21 days of treatment,group A underwent scar repair.In group B,the wound surface growth was poor,and the skin appendages were underdeveloped.All the wounds in group C were healed,and the epidermis,dermis and appendages became normal skin.Conclusions: 1.A full-thickness skin burn model of dogs can be successfully established by scald with 95℃ water 15 s.2.The bone marrow mesenchymal stem cell secreted factor was superior to the silver sulfadiazine group in treating full-thickness skin burns. |
