Citrus is an important economic crop in China,and it is also a mainly industry for increasing greenery and improving farmers'income in the mountainous areas of the south.It is also an important starting point for the government's precise poverty alleviation.After the great development of the industry,the prevention and control of epidemics of major diseases and insects became the primary task.In recent years,Citrus Huanglongbing?HLB?has occurred frequently in southern China,and the prevention and control become very serious.As an important pest in the young shoots of citrus,Diaphorina citri Kuwayama is the main natural insect vector that spread Citrus Huanglongbing.Diaphorina citri Kuwayama has attracted much attention in recent years.At present,the main control methods for Diaphorina citri Kuwayama still rely on chemical control,but the application of long-term insecticide has resulted in the resistance of Diaphorina citri Kuwayama to multiple insecticides.Therefore,how to effectively control Diaphorina citri Kuwayama while reducing the use of insecticides has become an urgent problem to be solved.Based on this,this dissertation takes the Diaphorina citri Kuwayama as the research object,and based on the platform of the National Navel Orange Engineering Research Center,on the basis of determining its sensitivity to 5 kinds of insecticides,studied the synergism of 3 synergists on the 5insecticides.Based on the bioassay test results of Diaphorina citri Kuwayama to the 5insecticides,thiamethoxam was selected to further study the effect of its sublethal dose on the biological characteristics of Diaphorina citri Kuwayama.Finally,we quantity the differences in protein in Diaphorina citri Kuwayama treated with thiamethoxam at two doses by using TMT.The main results are as follows:1 Diaphorina citri Kuwayama has different effects on different insecticides and synergistsThe Micro spot method was used to detect the sensitivity of indoors and fieldpopulations of Diaphorina citri Kuwayama to the same 5 insectcides.And then,the synergism of 3 synergists on these 5 insecticides was studied.The results are as follows:The susceptibility of the indoor population to the 5 insecticides is:Imidacloprid>Thiamethoxam>Dinotefuran>Fenpropathrin>Chlorpyrifos.There are no significant differences in the sensitivity levels of the 5 insectcides in two populations of Diaphorina citri Kuwayama;the synergism of 3 synergists on the 5 insectcides is generally PBO>TPP>DEM,and the synergism of different combinations are different.Among them,the combination of dinotefuran and PBO has the best synergistic effect,followed by the combination of fenpropathrin with PBO and TPP,and the combination with chlorpyrifos and DEM has the worst synergistic effect.2 Thiamethoxam sublethal dose affects biological characteristics of Diaphorina citri KuwayamaThe effect of thiamethoxam sublethal dose(LD20)on the biological characteristics of Diaphorina citri Kuwayama was studied by Micro spot method.The results showed that after the sublethal dose of thiamethoxam,the mortality rate of Diaphorina citri Kuwayama gradually increased with the treatment time increased.After 72 hours of insecticide treatment,the cumulative mortality rate reached 43.12%,and the cumulative mortality rate of the control was 25.42%.After 48 hours of dinsecticide treatment,compared with the control,the cumulative mortality rate of the treatment group was significantly higher.In terms of emergence rate,the treatment group was lower than the control,but there was no significant difference between the two groups in the three time periods.After thiamethoxam LD20 treatment,from the Diaphorina citri Kuwayama become adult until the females died completely,a total of 24 days of continuous statistical fecundity was recorded.Compared with the control,the fecundity amount of Diaphorina citri Kuwayama after the sublethal treatment of thiamethoxam are 591 fewer,but there is no significant difference between the two.3 Diaphorina citri Kuwayama respond to thiamethoxam-stressed proteinsWe quantity the differences in protein in Diaphorina citri Kuwayama treated with thiamethoxam LD50 and LD20 by using TMT technology.The results showed that among the 4521 proteins identified,GO annotated the most,2504.COG annotated to 1931,and KEGG annotated to 2142.In quantitative comparison analysis,after screening based on the principle of up-regulation?1.2 or down-regulation?0.83,P?0.05,it was found that compared with the control,the number of significantly differentially expressed proteins in LD50 and LD20 were 168 and 122.LD50 treated samples 83 proteins were up-regulated and 85 proteins were down-regulated;LD20 showed 84 proteins up-regulated and 38down-regulated.The function of these differentially expressed proteins is focused on Binding?catalytic activity?structural molecule activity?molecular transducer activity?transporter activity?antioxidant activity and enzyme regulator activity.
