Lignocellulase activity of seven strains which included pleurotus geesteranus,pleurotus nebrodensis,pleurotus ostreatus,pleurotus eryngii,pleurotus djamor,pleurotus citrinipileatus and pleurotus abalonus was determined.The enzyme production conditions and enzyme properties to those strains which good enzyme activity were further studied by liquid fermentation.The aim of this study was to select the excellent strains with high activity of degrading lignocellulosic enzyme in the pleurotus,optimize the culture conditions,improve the yield of degrading lignocellulosic enzyme,so as to improve the efficiency of degrading lignocellulosic enzyme,and lay the foundation for further development and application of degrading lignocellulosic.The following results were obtained through the experiment:(1)Laccase and cellulase activities in the tested strains were determined by ABTS and DNS method respectively.The result showed that the highest expression of laccase and cellulase was found in strain of pleurotus ostreatus which Laccase activity reached189.11U/mL,and cellulase activity reached 31.60U/mL.The other features of Pleurotus ostreatus was further research.(2)Effects of different carbon sources,nitrogen sources,metal ions and inoculation amount on degradation of lignocellulose of p.ostreatus were studied.The results showed that the relatively high laccase production formula of was 50 g/L glucose,8 g/L yeast extract,2 mmol/L copper ion and 4% inoculation.(3)The results of enzymatic characterization of laccase and cellulase showed that the temperature range of laccase and cellulase was 30 ~80?,the pH value range was3~8.The optimum reaction temperature of laccase was 37?.The stability of laccase was better within 60?,the optimum pH was 5,and the laccase was stable between 5and 7.The optimum temperature of cellulase is 45?,which is relatively stable when the temperature is less than 65?,the optimum pH is 5,and cellulase is relatively stable when the pH is between 4 and 6.
