Abstract/Summary:
As the largest metabolic organ in the body,liver plays an extremely important role in the occurrence and development of diseases.Therefore,protecting the liver is an indispensable measure to prevent livestock diseases.Therefore,this study further studied the protect effect of extract of Phytolaccae Radix and processed Phytolaccae Radix on CCl4-induced liver injury in chicken,and preliminarily explored its mechanism,which provided a basis for the clinical application of its.1.Study on the component changes of Phytolacca Radix before and after being processed with vinegar by FT-IR and HPLC:It was found that the main components of Phytolaccae Radix before and after the vinegar roasting basically did not change by FT-IR,but the content of KNO3 in processed Phytolaccae Radix decreased.The content of EsA in the extracts of processed Phytolaccae Radix was higher than that in the extracts of Phytolaccae Radix by HPLC.2.Preventive effect of the extracts of Phytolacca Radix and processed Phytolaccae Radix on CCl4-induced liver injury in chickens:180 1-day healthy chickens were randomly divided into 9 groups.The first group was the blank group,the second group was the model group,the third group was the drug control group(20 mg/kg of diphenyl diester);the fourth-sixth group was the low,medium and high dose group of the extracts of Phytolaccae Radix(0.4 mL/kg,1.2 mL/kg,2 mL/kg,respectively);the seventh-ninth group was the low,medium and high dose group of extracts of processed Phytolaccae Radix(0.4 mL/kg,1.2mL/kg and 2 mL/kg).On the 11th day,group 1-2 was given distilled water by gavage,and group 3-9 was given once a day for 5 days.On the15th day,chickens in group 2-9 were intraperitoneally injected with 10mL/kg of 25%carbon tetrachloride olive oil solution,while chickens in group 1 were intraperitoneally injected with the same amount of olive oil.After 48 hours,the chickens were killed.The liver index was calculated and the liver tissue was fixed.The expression of related enzymes,antioxidant factors,inflammatory factors and related proteins in serum was detected.The results showed that:(1)There was no significant difference in liver index among groups(P>0.05),but the liver index of chickens in model group was higher than that of other groups;(2)Compared with model group,the contents of AST,ALT,MDA,PEG2,IL-6,IL-1βand TNF-αin serum of chickens in different dose groups of the extracts of Phytolaccae Radix and processed Phytolaccae Radix significantly decreased(P<0.05).The contents of SOD,GSH-Px and CAT in serum were significantly increased(P<0.05);(3)the extracts of Phytolaccae Radix and processed Phytolaccae Radix could alleviate CCl4-induced liver injury;(4)Compared with model group,the extracts of Phytolaccae Radix and processed Phytolaccae Radix could significantly inhibit the expression of iNOS and COX-2 protein in liver tissue(P<0.05).3.Therapeutic experiment of the extracts of Phytolacca Radix and processed Phytolaccae Radix on CCl4-induced liver injury in chickens:180 1-day healthy chickens were randomly divided into 9 groups.The first group was the blank group,the second group was the model group,the third group was the drug control group(20 mg/kg of diphenyl diester);the fourth-sixth group was the low,medium and high dose group of Phytolaccae extracts(0.4 mL/kg,1.2 mL/kg,2 mL/kg,respectively);the seventh-ninth group was the low,medium and high dose group of processed Phytolaccae extract(0.4 mL/kg,1.2 mL/kg and 2 mL/kg).On the 15th and 18th day,the 2nd-9th groups were intraperitoneally injected with 10 mL/kg of 25%carbon tetrachloride olive oil solution,and the 1st group was intraperitoneally injected with the same amount of olive oil.Starting from the 16th day,group 1-2 was given distilled water,group 3-9was given corresponding drugs once a day for 5 days.21 days,the chickens were killed.The liver index was calculated and the liver tissue was fixed.The expression of related enzymes,antioxidant factors,inflammatory factors and related proteins in serum was detected.The results showed that:(1)There was no significant difference in liver index among groups(P>0.05),but the liver index of chickens in model group was higher than that of other groups;(2)Compared with model group,the contents of AST,ALT,MDA,PEG2,IL-6,IL-1βand TNF-αin serum of chickens in different dose groups of the extracts of Phytolacca Radix and processed Phytolaccae Radix significantly decreased(P<0.05).The contents of SOD,GSH-Px and CAT in serum were significantly increased(P<0.05);(3)the extracts of Phytolacca Radix and processed Phytolaccae Radix could alleviate CCl4-induced liver injury;(4)Compared with model group,the extracts of Phytolacca Radix and processed Phytolaccae Radix could significantly inhibit the expression of iNOS and COX-2protein in liver tissue(P<0.05).
