Melon(Cucumis melo L.)is an important commercially crop,and its growth and development is seriously affected by the land salinity.Previous studies have shown that Calcineurin-B-Like proteins play important roles in various biological stresses,especially salt stress in plants.However,there were few reports about Calcineurin-B-Like protein family in melon.In this study,in order to enrich the salt tolerance mechanism of melon,two Calcineurin-B-Like protein family genes CmCBL1 and CmCBL3 were isolated from the melon,and were investigated by methods of bioinformatics,subcellular location,tissue expression analysis and genetic transformation.The main results obtained are as follows:(1)Phylogenetic analysis showed that CmCBL1 and CmCBL3 belong to the Calcineurin-B-Like protein family.Confocal imaging of yellow fluorescent fusion protein(YFP)in transgenic tobacco(Nicotiana tabacum L.)leaves indicated that CmCBL1 and CmCBL3 were both located in the nucleus and plasma membrane.Transcript analysis showed CmCBL1mainly expressed in the leaves and stems of melon,and the relative expression level from high to low was:leaves,stems,roots.The relative expression level of CmCBL3 in melon had little difference among root,stem and leaf,but stems and leaves slightly higher than roots.Salt stress treatment will increase the expression of CmCBL1 and CmCBL3.(2)Overexpressing of CmCBL1 in wild type(WT)Arabidopsis thaliana can improve the salt tolerance of plants.The transgenic plants showed longer roots,higher maximum photochemical efficiency of PSII(Fv/Fm),higher K~+/Na~+ratio than the WT Arabidopsis thaliana under salt treatment.Under salt treatment,overexpressed CmCBL3 plants will reduce the germination rate of the seeds and reduce the total root length of the seedlings.However,the biomass decreased,but there was no significant difference in root length,plant height and expression between overexpressed CmCBL3 mature plants and wild ones under salt treatment.
