| Goat pox,also known as "goat smallpox," is an acute,febrile,sheep-induced Contact infectious diseases are listed by the World Organisation for Animal Health(OIE)as a notifiable infectious disease.The characteristic clinical lesions of sheep pox are the specific rash and pimples that appear on the skin and mucosal surface of the affected sheep,and then turn into blisters,pustules,or scabs.The temperature of the affected sheep usually increases in the early stages.Sheep pox virus mainly infects sheep and goats,and some wild ruminants can also be infected.In addition,the virus can infect humans.Sheep infectious impetigo(commonly known as sheep aphtha),which has many similarities to the clinical symptoms and pathogenesis of sheep pox,is also a common infectious disease in sheep.Both are often misdiagnosed.Sheep’s aphtha is a highly contact,epithelial infectious disease caused by sheep infectious pustular virus(ORFV).In addition to sheep,ORFV can infect a variety of other ruminants,some aquatic animals,and humans.Goat pox and aphthous ulcers are both important zoonotic infectious diseases.The outbreak of the disease will not only bring serious economic losses to the breeding industry,but may also threaten the health of farmers,shepherds and veterinarians.Therefore,the research on the correct diagnosis and treatment of similar clinical cases can provide a certain guarantee for the healthy development of the sheep industry.In October 2018,60 small-tailed cold sheep raised by a sheep farmer in Nong’an,Jilin Province became ill.The temperature of the sheep in the early stages increased,the spirit was depressed,and the body was thin.Among them,four lambs about one month old had proliferative lesions such as pimples and pimples at different levels on the hairless or hairless areas such as the lips,tongue,eyes,and tail roots.At last thescabs were broken and some sheep were cured.According to the incidence and clinical symptoms,it is presumed to be caused by sheep pox virus(SPPV)infection.In order to further confirm the diagnosis,based on a rapid,accurate and effective method for the differential detection of sheep pox virus and sheep pox virus,this study carried out systematic experiments on clinical suspected sheep pox cases from the perspective of pathology and etiology.Laboratory testing and analysis,as well as the isolation and identification of the virus,finally confirmed the disease.In this paper,a single PCR method for the identification of sheep poxvirus and sheep mouth sore virus was successfully established in a suspected case of sheep poxvirus infection.Then,by optimizing the conditions of primer concentration and annealing temperature in the PCR reaction system,as well as the study of specificity and sensitivity,the double p-pcr of sheep poxvirus and sheep mouth sore virus with good specificity and high sensitivity was successfully established Differential diagnosis of PCR.At the same time,a suspected case of sheep pox virus infection was diagnosed.According to the established diagnosis method and laboratory analysis of pathology and etiology,the clinical case was confirmed as sheep pox.In order to further study the virus strain,we isolated and identified the virus and analyzed its molecular characteristics by inoculating sheep pox virus solution into oftu cells,observing the cytopathy and electron microscopy negative staining,TCID50 determination,and molecular genetic evolution analysis.The results showed that the Double PCR detection method of sheep pox virus and sheep mouth sore virus was successfully established in this experiment,which had good specificity and high sensitivity.In addition,through systematic laboratory analysis and virus isolation and identification methods,this study confirmed a clinical case of suspected sheep pox virus infection,and successfully isolated a sheep pox virus.In this study,multiple laboratory testing methods were used to confirm the diagnosis of suspected sheep pox cases,and laboratory diagnostic techniques were established to identify sheep pox and sheep infectious impetigo,and wild strains were isolated.It has laid a foundation for the follow-up research related to the diagnosis and prevention of sheep pox. |
