Melon is loved by everyone because of its sweetness.The fruit texture of different varieties of melons varies greatly and has a variety of different tastes.Previously,the main breeding goal of melon breeding was to cultivate melon varieties with high sugar content,high yield,resistance to diseases and insect pests,etc.,and less attention was paid to the texture characteristics of melons with crisp taste.At present,melon accumulations with crisp taste are welcomed by the consumer market.In this experiment,five kinds of thick-skinned melon materials with significant differences in taste were used to explore the changes in the cell wall material of melon pulp at different development stages,the changes in cell wall enzyme activity related to texture,and the process of cell wall expansion enzyme genes during different stages of fruit development,ripening and softening The changes in expression level were measured by TPA and puncture methods.The changes of texture parameters of muskmelon fruit in different periods and the observation of pulp cell morphology.Crispy melons are the current breeding direction.This experiment studied the internal factors of the texture differences of the five melon materials,providing theoretical guidance for the breeding of crisp melon melons and exploring the mechanism of melon texture formation.By constructing separate populations to analyze the genetic inheritance laws and applying the BSA-seq method to initially locate the genetic inheritance traits,it provides a theoretical basis for the molecular marker-assisted breeding,shortens the breeding cycle,improves the breeding efficiency,and breeds new crispy melon varieties.The main conclusions as follows:1.This experiment studied the changes of pulp texture and related enzyme activities during the development of 5 different types of muskmelon fruits.Studies have shown that the 35 th day after flowering is a critical period for the formation of the texture of soft flesh melons.Cell size parameters are significantly correlated with pulp texture parameters.During this period,the cell area and gap of soft flesh melon increased,and the flesh cells were loosely arranged.The texture parameters of soft flesh melon and crisp flesh melon were significantly different.The activity of four cell wall enzymes of crisp flesh melon fruit was lower than soft flesh Type melon.2.Different materials of muskmelon have different changes in the content of the cell wall during various stages of fruit development.From 15 to 40 days after flowering,the CSP content generally showed a downward trend,and the WSP content generally showed an upward trend.The CSP content of soft-fleshed melons and melons was higher than that of crisp-fleshed melons,and the CSP and WSP contents were higher than ISP.3.The pulp cell morphology parameters were significantly correlated with TPA hardness and puncture hardness.4.Studies have shown that 35 d after flowering is a critical period for cell wall expansion enzyme gene expression.During this period,the relative expression of the cell wall expansion enzyme genes(Cm EXP3,Cm EXP5,Cm EXP9)of soft-skinned melon 'NSL' was the highest,which was significantly higher than that of 'XZM'.5.This study used the BSA method to locate the melon crispness gene.The BSA method initially locates the skin color related genes in the 26.64-31.33 Mb and 22.41-25.24 Mb regions of chromosome 03 and chromosome 09,respectively,with a total length of 7.60 Mb and contains a total of 1,060 genes,including non-synonymous mutation SNP There are 16 genes in total.
