Cattle-yak is an interspecies F1 hybrid of a cross between a yak and Bos Taurus.It has obvious hybrid advantages,and has outstanding milk production,meat production,body size,and service ability.Protein is the performer of life activities,and it participates in life.The entire process involves the expression of a large number of genes and network-based regulation,which is characterized by diversity and complexity.Understanding the regulatory relationships between differential proteins between muscle growth and development and fat deposition is of great significance for yak and Cattle-yak in breeding and breeding work.Therefore,in this study,TMT technology and bioinformatics methods were used to screen differentially expressed proteins and pathways of yak and Cattle-yak in Longissimus dorsi and subcutaneous fat.In the experiment,three adult healthy female yak and female Cattle-yak were collected.After slaughter,the Longissimus dorsi and subcutaneous fat adipose tissue were collected,and the total protein of each tissue was extracted.TMT technology was used to identify differentially expressed proteins.After reagent labeling,mass spectrometry identification and protein database search were performed,and bioinformatics analysis was performed.The main research contents and results are as follows:1.The results showed that when 0.06?FC?1.4 and p-value?0.05,there were 37 proteins that were significantly different in subcutaneous fat;113 proteins were significantly different in the longissimus dorsi.2.Through the analysis of go significance,it was found that in subcutaneous fat tissue,differential proteins were enriched in 64 GO terms,mainly in extracellular matrix structural components,oxidoreductase activity,fibrin complex and oxidationreduction process;in Longissimus dorsi,differential proteins were enriched in 128 GO terms,mainly in catalytic activity,mitochondrial inner membrane,metabolic process and ATP Metabolic engineering.3.The results of KEGG signal pathway analysis showed that in subcutaneous fat tissue,there were 66 KEGG signal pathways,including protein digestion and absorption,steroid hormone synthesis and receptor interaction.In the longissimus dorsi,there are 77 KEGG signaling pathways,including butyrate metabolism,carbon metabolism and citric acid cycle.4.The results of protein interaction analysis showed that in subcutaneous fat,proteins such as tendon protein X precursor,flavoamine oxidase B,acyl coenzyme A synthetase ACSM1 and metalloproteinase inhibitor 2 precursor had higher network connectivity,which might play an important role in fat deposition mechanism.Glutathione transferase M1,NADH isocitrate dehydrogenase and prostaglandin reductase 1 may play an important role in the development of muscle.In this study,a variety of differentially expressed proteins related to muscle growth and fat deposition were screened,and the protein expression in the longissimus dorsi and subcutaneous fat tissues of yaks and Cattle-yak was more comprehensively understood,which laid a good foundation for further study on the mechanism of the longissimus dorsi and subcutaneous fat tissues of yaks and Cattle-yak.
