Arsenic is a metal-like pollutant widely present in the environment,mainly in the form of inorganic arsenic and organic arsenic in the environment.Its sources mainly include industrial production,mining of arsenic-containing minerals,and use of arsenic-containing products.After low concentration of arsenic enters the organism,it can promote the growth of the organism.At high concentration,it will cause the toxic effect of the organism and harm the growth and development of the organism.With the large-scale use of arsenic-containing products such as feed,fertilizers,herbicides,wood preservatives,etc.in animal husbandry and agriculture,the enrichment of arsenic in organisms has been accelerated,which further promotes their harm to organisms.Dimethyl arsenic acid is a kind of organic arsenic.It is the main metabolite of inorganic arsenic that undergoes methylation metabolism after entering the organism.Because of its strong toxicity,it has been classified as a carcinogen by the World Health Organization and is an important component of arsenic pollution.At present,there are two main research directions for arsenic: one is environmental arsenic pollution assessment,and the other is the impact of inorganic arsenic on organisms.At present,research on the effect of organic arsenic dimethyl arsenic acid on mammals is still very rare.Liaoning Cashmere Goat is my country's unique meat cashmere dual-use sheep species.Its cashmere output is high and the quality is good.It has been listed as a key protected resource by the state and is prohibited from export.With the continuous deterioration of arsenic pollution,the harm trend of arsenic on Liaoning cashmere goats is becoming more and more serious,and it also has a potential impact on the yield and quality of important product hair.Therefore,it is of great significance to study the effect of dimethyl arsenic acid on the skin fibroblasts of Liaoning cashmere goats.In this study,immunofluorescence and Western Blot technology were used to detect the expression levels of keratin and collagen in Liaoning cashmere goat skin fibroblasts treated with dimethylarsenic acid.Using q PCR technology to study the expression of target genes related to hair growth in fibroblasts treated with high concentration of dimethylarsenic acid,and to detect the regulation of related pathways p38 MAPK and NF-? B.Through the combined action of dimethylarsenic acid and pathway inhibitors,observe the expression of target genes and pathway regulation.The purpose of this paper is to investigate the changes of keratin and collagen expression in skin fibroblasts of Liaoning cashmere goats treated with dimethylarsenic acid,and to observe the effects of the m RNA expression of keratin-related target genes KAP26.1,K26,KAP11.1,KAP8.2,KAP7.1,KAP6.2 and collagen-related genesCOL1A1 and COL1A2,the aim is to regulate the growth of cashmere hair.And explore whether p38 MAPK,NF-? B signaling pathway is involved in mediating this regulatory process.Through research,it was found that 0.80mmol/L dimethylarsenic acid can promote the expression of collagen in fibroblasts,but it has almost no effect on the expression of keratin.35.00 mmol/L dimethyl arsenic acid was able to inhibit the expression of collagen in fibroblasts,but significantly promoted the expression of keratin.However,both low and high concentrations of dimethylarsenic acid can promote the expression and inhibition of keratin and related protein target genes KAP26.1,K26,KAP11.1,KAP8.2,KAP7.1,KAP6.2m RNA.Type I collagen egg gene COL1A1,COL1A2 m RNA expression.Both p38 MAPK and NFk B signaling pathways are activated to varying degrees in response.After analysis,p38 MAPK signaling pathway may be involved in dimethyl arsenic acid on Liaoning cashmere goat skin fibroblast keratin and its associated protein gene KAP26.1,K26,KAP11.1,KAP8.2,KAP7.1,KAP6.2 expression regulation.The NF-k B signaling pathway may be involved in the regulation of the expression of collagen-related genes COL1A1 and COL1A2 by dimethylarsenic acid.This provides a theoretical basis for revealing that organic arsenic dimethyl arsenic acid can participate in regulating the growth of Liaoning cashmere wool,and lays a foundation for the study of the specific mechanism of regulating.
