In this experiment,the steam explosion pretreatment technology was used to process Caragana korshinskii in different growth periods,and the dominant strains of yeast,Bacillus subtilis and lactobacillus with fiber degradation ability were screened,separated and purified by our laboratory.After optimization of fermentation parameters,they were used as composite agents to ferment Caragana powder,so as to reduce its fiber components and improve feed quality.The purpose of this study was to explore the steam explosion The effect of fermentation and double treatment on the fiber components of Caragana korshinskii.Experiment 1: the effect of steam explosion on the degradation of Caragana fiber componentsThe purpose of this experiment is to study the effect of steam explosion on the degradation of Caragana fiber.In the flowering stage(April),fruiting stage(July)and defoliation stage(November),the whole Caragana korshinskii was collected to determine its conventional nutrients;the content of neutral washing fiber,acid washing fiber,lignin,hemicellulose and cellulose were determined by steam explosion pretreatment at different pressures(0.8MPa,1.1MPa and 1.4mpa)for 3min.The results showed that: 1)the feed value of Caragana korshinskii was higher from the content of main nutrients.2)Compared with the control group,the content of neutral washing fiber,acid washing fiber,lignin and cellulose increased first and then decreased(P < 0.05),hemicellulose decreased linearly(P < 0.01),and neutral washing soluble matter decreased first and then increased(P < 0.05)with the increase of steam explosion pressure.The results showed that the nutrition value of Caragana korshinskii was the highest when it was planted in flat stubble,and the fiber component of Caragana korshinskii could be significantly reduced by steam explosion,which was beneficial to its feeding value.Experiment 2: optimization of fermentation conditions of Caragana by complex bacteriaIn this experiment,the amount of compound bacteria,the proportion of bacteria inoculated and the water content were taken as the factors of fermentation parameters.Orthogonal design was used to optimize the fermentation parameters of Caragana korshinskii with NDF,ADF and lignin degradation effect as reference.The results showed that: 1)the degree of fibrosis of Caragana korshinskii in defoliation stage was the highest than that in other growth stages,and the fiber components of Caragana korshinskii in defoliation stage could be significantly reduced by the fermentation of complex bacteria.2)Taking NDF,ADF and lignin degradation as reference,the fermentation effect of test group 7 was significantly better than that of other test groups.3)Taking hemicellulose and cellulose as reference,the experimental group 4(dosage: 5%;inoculation ratio: Lactobacillus: yeast: Bacillus subtilis = 1:3:5;water content: 40%)was significantly better than other experimental groups.The results showed that the fermentation parameters of Caragana microphylla by complex bacteria were as follows: the addition amount of bacteria: 10%;the inoculation proportion of bacteria: Lactobacillus: yeast: Bacillus subtilis = 3:5:1;the water content was 40%,which significantly degraded the fiber components of Caragana microphylla.Effect of steam explosion,compound bacteria fermentation and double treatment on fiber components of CaraganaThe purpose of this experiment is to explore the effects of steam explosion,fermentation with complex bacteria and their dual treatment on the components of Caragana fiber.Steam explosion pretreatment with different pressure(0.8 MPa,1.1 MPa and 1.4 mpa)was carried out for 3min,and then 10% mixed culture medium(Lactobacillus: yeast: Bacillus subtilis = 3:5:1)was inoculated for solid-state fermentation.The content of neutral detergent fiber,acid detergent fiber,lignin,hemicellulose and cellulose,and the activity of carboxymethylcellulose and xylanase in fermentation substrate were determined.The results showed that: 1)compared with the same period before fermentation,the content of neutral washing fiber(P < 0.01),acid washing fiber(P < 0.01),hemicellulose(P < 0.05)and cellulose(P < 0.01)decreased significantly after fermentation,and the content of neutral washing soluble substance(P < 0.01)increased significantly.2)Compared with the control group,the content of neutral washing fiber(P < 0.001),acid washing fiber(P < 0.05),hemicellulose(P < 0.001)and cellulose(P < 0.01)decreased significantly,and the content of neutral washing soluble substance(P < 0.01)increased significantly with the increase of bursting pressure The lignin content of Caragana korshinskii(P < 0.05)decreased first and then increased in the flowering and fruiting stage,and decreased significantly in the deciduous stage(P < 0.01).3)Compared with the control group,the activity of carboxymethylcellulase(P < 0.01)decreased first and then increased with the increase of steam pressure,while the activity of carboxymethylcellulase(P < 0.01)increased significantly at the fruiting stage,and the activity of xylanase(P < 0.01)in the fermentation medium of Caragana during the flowering,fruiting and defoliation stages increased with the increase of steam pressure The increasing trend is linear.The results showed that steam explosion,compound bacteria fermentation and double treatment could significantly reduce the fiber components of Caragana,and the effect of compound bacteria fermentation was better after steam explosion.
