Sorghum is one of the most important food crops in the world.The endosperm accounts for 80% of the kernel and provides the most important place for nutrient storage.Gene imprinting is a classic epigenetic phenomenon,its performance for the allele-specific expression of a class of genes is dependent on the parental origin of control,mainly occurring in the endosperm,which plays an important role in endosperm development.Gene imprinting includes not only the protein-coding genes,also including non-coding RNA.Long non-coding RNA(lncRNA)are a class of length greater than 200 bp,and does not have the ability to transcripts encoding a protein,which plays an important role in many biological processes of growth and development and molecular regulation.Currently,lncRNA has been excavated and identified in the genomes of many species,but with a mark of identification of lncRNA state but rarely reported.In this review,we collected deep RNA-seq of endosperm isolated 14 days after pollination(DAP)from inbred sorghum BT × 623 and Y49 reciprocal sorghum F1 hybrids.lncRNA and imprinted lncRNA in the endosperm of sorghum on the 14 th day after pollination were identified in the genome by using the method of Genome-guided.The characteristics of lncRNA and imprinted lncRNA and their expression in different tissues were also analyzed.The main results are as follows:1.According to a feature transcript length,protein coding capability and expression abundance,using strict screening criteria to identify the genome-wide of potential lncRNA in Sorghum,4388 lncRNA loci(5310 transcripts)were identified.From the identified lncRNA a total of 44 imprinted lncRNA were screened,including 44 expressed preference for maternally imprinted lncRNA and not identified to imprinted lncRNA expression of paternal.2.According to the location of lncRNA and annotated genes,we classified lncRNA,among which 29% were intergenic lncRNA,61% were genetic overlapped lncRNA,and 10% were antisense lncRNA.Identified imprinted lncRNA were divided into 27 long non coding RNAs which landed in the intergenic region,14 long-chain non coding RNAs which overlapped with genes and 14 antisense long-chain non coding RNAs.3.The distribution of lncRNA in genome is uniform,but the average density of chromosome 1 is higher than that of other chromosomes.The distribution of imprinted lncRNA in the genome is more uniform.4.The average number of exons was 3 and the average length was 1017 bp.The average number of exons was 3 and the average length was 1007 bp.5.The tissue-specific expression of lncRNA is characterized by high abundance in specific tissues and almost no expression in other tissues,while the constitutive expression of lncRNA is generally characterized by low abundance in many tissues.73% of imprinted lncRNA showed strong tissue expression specificity in endosperm.In conclusion,this study is not only to provide a reference for assisted breeding high-yielding sorghum,also can further enrich people's awareness of the grasses of the imprinted lncRNA.
