Phalaenopsis plants are very popular because of their high ornamental value,and the annual demand for Phalaenopsis in the flower market is huge.At present,the industrial production of Phalaenopsis mostly adopts asexual reproduction group cultivation seedling technology,but the survival rate of tissue culture seedlings is low,the adaptability is poor,the offspring traits are easy to degenerate,and new traits are not easy to produce.This study took a Phalaenopsis aphrodite cv.MF as the research object,and the symbiotic system was established with the mycorrhizal fungi isolated from the wild Oreorchis patens and Liparis japonica in northeast China,and two fungi that significantly promoted seedling growth were screened;To further improve the Phalaenopsis and mycorrhizal fungal symbiosis system,provide new technology for mycorrhizal seedlings for the industrial production of Phalaenopsis,and make up for the shortcomings of the current tissue culture technology,has great potential application value for the industrial production of orchids.The main research contents and results are as follows:(1)Oreorchis patens was collected from Changbai Mountains in Jilin,and isolation of mycorrhizal fungi from the roots.The obtained mycorrhizal fungi were identified by ITS sequence.The results showed that we isolated three mycorrhizal fungi from the root of Oreorchis patens,cbssl-1 was classified as Tulasnella,Gen Bank sequence number was MH050440;cbssl-2 was classified as Chaetomium,Gen Bank sequence number was KY630740;cbssl-3 was classified as Fusarium,Gen Bank sequence number was KY630741.(2)Cbssl-1 and six Tulasnella fungi which isolated from the roots of wild Liparis japonica in Northeast of China established a symbiotic system with Phalaenopsis seeds and protocorms.The results showed that none of the seven fungi we selected promoted Phalaenopsis seeds.However,both Fungi 114 and Fungi 2 promoted the growth of the protocorm of Phalaenopsis.Compared with the control group,the growth promotion effect of Fungi 114 on the protocorms of Phalaenopsis was the most significant,and the growth rates of dry weight and fresh weight and leaf width reached 229.63 %,282.02 %,and 154.78 %.(3)The screened beneficial Fungi 2 and 114 were cultured in vitro to investigate their biological characteristics,and researched the growth differences of Fungi 2 and 114 under different carbon sources,nitrogen sources,p H,and temperature.The results showed that the most suitable carbon source for Fungi 2 was maltose,the most suitable nitrogen source was potassium nitrate,the most suitable temperature was 30 ?,and the most suitable p H was 7;the optimum carbon source for fungi 144 was lactose and the optimum nitrogen was ammonium nitrate,the optimum temperature was 25 ?,and the optimum p H was 6.
