?Objective?In the recent years,cultivated products has replaced wild products as the main sources of Chinese herbal medicines,and the use of plant growth regulators(PGRs)in the cultivation of Chinese medicines were frequently reported.PGRs are pesticides that can be used to increase crop production,as well as improve the quality of crops.However,some PGRs also bring toxicity to human health and the environment.Chinese Pharmacopoeia(Ch P)has set the limit for some organochloride pesticide residues in Chinese medicines but still has no limit for those of PGRs.Moreover,the standard methods for determination of PGR residues in National Food Safety Standard are mostly complicated and time-consuming single-residue methods.Our study's purpose was to develop a rapid,easy,and accurate high-throughput method for the simultaneous determination of multiple PGR residues in Chinese medicines,so as to provide reference for future development of PGR residue regulations in Chinese medicines.?Method?Codonopsis Radix(Dangshen)and Notoginseng Radix et Rhizoma(Sanqi)were selected as subjects of the study.The analysis of PGR residues was performed using UPLC-MS/MS technique,whereas sample preparation procedures were optimized based on Qu ECh ERS method.Electrospray ionisation(ESI)and a triple quadrupole(Qq Q)analyser in scheduled multiple reaction monitoring mode were used for mass spectrometry analysis.0.05% formic acid 5 m M ammonium formate in methanol and water were selected as mobile phase.For sample preparation,acetronitrile was used as extraction solvent and sodium citrate buffer was used as extraction salt.Finally,the method validation was performed following European Commission Guidance Document on Analytical Quality Control and Method Validation Procedures for Pesticide Residues and Analysis in Food and Feed(SANTE/11813/2017)?Results? The developed method was validated in accordance with SANTE/11813/2017 guideline,showing good linearity of the calibration curves,acceptable recovery and RSD of 69.1-119.8% and 0.1-19.8% for Dangshen;73.6-112.0% and 0.4-19.5% for Sanqi.LOQ were 3-20 ?g/kg and 3-10 ?g/kg for Dangshen and Sanqi,respectively.The method was also applied to determine PGR residue levels in 35 batches of Dangshen and 60 batches of Sanqi samples.In Dangshen 10 PGR residues were detected,whereas in Sanqi 9 of which were detected.?Conclusion?A multiple-residue method for the simultaneous determination of 39 PGR residues in Dangshen and Sanqi was successfully developed and validated following SANTE/11813/2017 method validation criteria,and was successfully applied in the analysis of commercial products.
