Objective:Detection of PITX3 Methylation levels in Peripheral Blood of Patients with Parkinson's Disease by Methylation-Sensitive High-Resolution melting Curve analysis,Compared with normal people of the same age group,At the same time,the significance of the new method is discussed.Methods:The peripheral blood is the object of study in 60 cases of Parkinson's disease and 20 cases of normal subjects,after extracting DNA and bisulfite treatment,methylation sensitivity of high resolution curve(MS-HRM)was used to detect PITX3 gene methylation.The methylation degree of patients and normal subjects and the degree of methylation in different hoehn-yahr stages were analyzed.Results:Comparison of basic clinical data,the results showed that there was no statistical difference between the age and gender of the two groups,and P > 0.05.41 of Parkinson's patients have a loss of olfaction(68.33%),34 of Parkinson's patients have sleep disorders(56.67%),41 of Parkinson's patients have constipation(68.33%).There were statistically significant differences between the two groups in olfaction,sleep disorder and constipation.The experimental group compared with control group.two groups of methylation level by using two independent sample t-test,the results showed that the difference in the two groups(t = 6.109,P < 0.05).The degree of methylation in the case group was higher than that in the control group.the methylation degree of the early stage and the middle and late stage(37 cases)of Parkinson's disease(23 cases),t-test was adopted,with no statistical difference(t=0.016,P>0.05).Conclusion:The results of our study showed that compared with normal controls,pitx3 gene promoter in Parkinson's disease was more methylated.in patients with parkinson's patients.In hoehn-yahr,There was no significant difference between early and late stage methylation.
