Background and objectives The Human glioma are the most common type of nervous system tumors,accounting for 50% to 60% of intracranial tumors.The adult morbidity of glioma is 6/10 million,and 5-year survival rate is 20% to 30%.At this stage,the treatment of glioma is based on surgical resection,combined with radiotherapy,chemotherapy,and biological therapy,including more than 80 gene therapy programs involving nearly 10,000 patients.Despite the treatment of glioma have been made great improvements,the results have not changed much compared with the 5-year survival rate of 17%-24% of the statistics five years ago,and the treatment effect is not satisfactory.To explore of the occurrence and development mechanism of gliomas in-depth exploration of new effective therapeutic targets has become the focus of current research.Interleukin-8(IL-8),also known as CXCL8,was first discovered as a chemokine that can chemoattract neutrophils and play an important role in many inflammatory response diseases.IL-8 can be secreted by a variety of cells,such as monocytes,macrophages,epithelial cells,endothelial cells,and the like,.IT can exert its biological effects mainly by binding to its receptors CXCR1 and CXCR2.With further understanding of the function of IL-8,it has been found that IL-8 is also a functionally diverse cytokine,which is not only related to the occurrence of inflammatory diseases,but also plays an important role in the progress of tumor.Studies have shown that tumor-derived IL-8 promotes the proliferation and invasion of tumor cells through autocrine and paracrine methods,promotes epithelial-mesenchymal transition,and promotes angiogenes in tumor to promoting the occurrence and development of tumors.In glioma,IL-8 is highly expressed,but the relevant analysis of clinically parameters with IL-8 in glioma patients,and the mechanism by which IL-8 promotes tumor progression are unclear.Therefore,the main task of this study is to analyze the correlation between IL-8 expression and clinical parameters in glioma patients and to explore the possible mechanism of IL-8 in promoting glioma progression.Methods This study included clinical data and m RNA microarray data from 307 glioma patients from the Chinese Glioma Genome Atlas(CGGA)database(http://www,cgga.org.cn).We downloads m RNA expression RNASEq V2 data from glioma datasets The Cancer Genome Atlas(TCGA)database(http://tcag-data.nci.nih.gov/tcga/),and preprocesses these date while downloading clinical data from patients with glioma.Accroding these data,IL-8 expression and clinical relevance are analysised.From October 2016 to October 2017,64 patients with primary brain glioma who were treated by brain surgery at the First Affiliated Hospital of Zhengzhou University were selected.All patients were pathologically diagnosed as glioma.All patients signed a written informed consent form and did not receive any chemotherapy or radiation therapy before admission.This study was approved by the Ethics Committee of the First Affiliated Hospital of Zhengzhou University.RT-PCR was used to detect the expression of IL-8 in different grades of glioma patients.Results 1.Compared with low-grade glioma,IL-8 is highly expressed in high-grade glioma.2.The expression of IL-8 is higher in malignant glioma with a higher degree of malignancy than that with low malignancy.3.IL-8 is expressed in IDH1 wild-type glioma with a higher degree of malignancy than IDH1 mutants with a lower degree of malignancy.4.IL-8 expression levels are significantly associated with glioma grade,IDHmutations,and tumor types in patients.5.Patients with lower IL-8 expression have better prognosis than patients with high IL-8 expression.6.IL-8 is an independent factor affecting the prognosis of patients with glioma.7.IL-8 may influence the prognosis of glioma patients through signaling pathways such as angiogenesis,cell adhesion,platelet degranulation,and cytokine secretion.Conclusion IL-8 is highly expressed in highly malignant gliomas,and the high expression of IL-8 predicts poor prognosis of patients.IL-8 is an independent factor that affects the prognosis of glioma patients.IL-8 may influence the prognosis of glioma patients through signaling pathways such as angiogenesis,cell adhesion,platelet degranulation,and cytokine secretion.
