Blood-brain barrier requires a tightly regulated local environment for cells to function.And is responsible for maintaining homeostasis of the brain by regulating the chemical environment,immune cell transport,and the entry of xenobiotics.In recent study,scientists notice the blood-brain barrier permeability has increased in Neurodegenerative disease like Alzheimer's disease and Parkinson's disease.It's necessary to establish a Blood-brain barrier in vitro which can represent the endothelium systemInsulin plays a critical role in glucose homeostasis by regulating the balance between glucose production by the liver and glucose uptake by muscle and adipose tissues.Disruption of insulin signaling,however,makes neurons more vulnerable to metabolic stress,thus accelerating neuronal dysfunction.In recent study,scientists notice the blood-brain barrier(BBB)permeability has increased in Alzheimer's disease.It is showed that the BBB has an important relationship with the Alzheimer's diseases.Decreases in insulin levels in CSF in patients with Alzheimer's disease,suggesting that may be related to insulin and neurodegenerative diseases.In this experiment,we choose different concentrations of insulin interferencing the hCMEC/D3 cells which act as a BBB model in vitro.The effect of insulin action on the permeability of blood brain barrier model was tested by detecting the expression of mRNA,ITGB1 and(FHL2,CDH5)and transport carrier(ABCB1,ANAX2)as well as the expression of inflammatory cytokines.The model of the BBB has nearly doubled the rate of electrical resistance,and it is higher than that of the control group from 20~30?·cm~2,which indicates that insulin can accelerate the formation of the complete model.Tight junction proteins(FHL2,ITGB1 and CDH5),transporters(ABCB1,ANAX2)mRNA expression was significantly increased.No changes in the expression of inflammatory cytokines mRNA,Insulin may decrease the permeability of blood brain barrier by promoting cell tight junction.Insulin on BBB integrity model establishment,and closely connected with an important role,so to maintain a certain level of insulin facilitates the establishment conforms to the endothelial brain micro environment in vitro BBB model.Therefore,changes in the level of insulin for research on Alzheimer's disease may have an important significance.
