| Objective: To study the effect of three different types of anti adhesion materials of M1,M2 and M3 to prevent postoperative abdominal adhesion,and to explore the possible mechanism of action in order to find more effective measures to prevent adhesion and adhesion of abdominal adhesions.Methods: 80 New Zealand rabbits were randomly divided into 4groups,including model control group,M1 material group,M2 material group and M3 material group,20 in each group.The model of intraperitoneal adhesion was made.The model control group of New Zealand rabbits did not do further treatment.The relative contact of the abdominal wall group defect and the cecal injury was placed into the abdominal cavity.The M1,M2 and M3 materials groups were sutured at the four corners of the injured abdominal wall and the damaged intestinal canal to fix the experimental materials of2cmx2.5cm size,and the abdominal wall group defect and the cecum loss The injuries were placed into the abdominal cavity relative to the contact.The degree of adhesion was observed by naked eyes at 7 and30 days after operation.HE staining and Mason tricolor staining,TGF-β1 immunization and Collagen I immunohistochemistry were taken to observe the morphological structure and cell growth of the material by scanning electron microscope.Statistical processing: all data SPSS 20 software is completed.P<0.05 has statistical significance.Result:The difference of M1 material group,M3 material group and model control group was statistically significant(P<0.05)7 days after the 1.adhesion degree score(P<0.05).The difference between the M2 group and the M1 material group was statistically significant(P<0.05),and the difference between the M3 material group and the M2 material group was statistically significant(P<0.05).30 days after operation,compared with model control group,the difference between M1 material group and M3 material group was statistically significant(P<0.05).2.HE staining and measurement results indicated that there was a significant difference between the M1 material group and the M2 Material Group on the 7 day after operation compared with the model control group(P<0.05).The difference between M1 material group and M2 material group was statistically significant compared with model control group(P<0.05).3.TGF-β1 immunohistochemical staining results showed that the expression of TGF-β1 in the control group was not significantly different from that in the M1 group 7 days after the operation,and the expression level of TGF-β1 in group M2 and M3 was significantly higher than that in the control group and the M1 group.On the 30 day after operation,there was no significant difference in the expression of TGF-β1 between the control group and the M1 group.The expression level of TGF-beta 1 in the M1 group and the M2 group was significantly lower than that in the control group and M3 group.On the 7 day after operation,the expression of Collagen I was significantly increased in the control group.There was no significant difference in the expression level between group M2 and M3,and the expression level of Collagen I in group M1 was significantly lower than that in the above three groups.On the 30 day after operation,the expression level of Collagen I in group M2 was significantly lower than that in the other three groups,and the expression level of Collagen I in group M3 was significantly higher than that in the other three groups.4.scanning electron microscopy(SEM)that M2 is a polymer electrospun porous membrane with inhibition of fibroblast proliferation,with poor strength and flexibility,M3 is a simple degrading polyester flow film with excellent strength and flexibility;M1 is a composite membrane of the above two,that is,a porous membrane junction that inhibits the proliferation of fibroblasts at the same time.The strength and elongation at break of the structure are two.M3 cast film and M1 cast film surface have nano scale depression,not completely smooth surface.Conclusion:(1)M1,M2 and M3 can inhibit the occurrence of intraperitoneal adhesion to a certain extent.The effect of M1 anti adhesion material is the most ideal,and it is a potential anti adhesion material with potential for clinical application.(2)the higher the expression level of TGF-beta 1,the more serious the adhesion was.TGF-beta 1 is closely related to the formation of intestinal adhesion.(3)physical barrier function and drug action play a synergistic role in anti adhesion process,which provides a new research direction for prevention and control of adhesion. |
