Objective:In order to explore the effect and mechanism of Tongfengxiao 2 on acute gouty arthritis,this experiment observe the circumference and pathological changes of rats'joints,the content of IL-1?IL-6 and TNF-? in rats' serum and the content of VCAM-1?ICAM-1 and NF-?B in rats'joints,after the rat model of acute gouty arthritis was replicated while intragastric administration of Tongfengxiao 2.Method:84 clean grade SD male rats,with the weight in(180±20)g,can be divide into 6 groups:blank group,model group,western medicine group,high dose group of Tongfengxiao 2,middle dose group of Tongfengxiao 2 and low dose group of Tongfengxiao 2,and each group has 14 rats.After water extraction of Chinese herbal compound concentration,dissolved in physiological saline make into low,middle and high dose group of Tongfengxiao 2;colchicine tablets dissolved in physiological saline into western medicine group.After 18 days of dosing cycle,except the blank group,the rats were selected to use No.4 pinhead to inject 0.2ml sodium urate solution into its right ankle joint,with the other side of joint capsule blow up to be the standard,to make the gout model successful.After 4 days,anesthetized the rats,then use the blood taking needle to collect blood from the abdominal aorta,3000 r/min centrifuged the blood into serum for 10min,stored at-20 ?.Use the ELISA method to measure the content of IL-1??IL-6 and TNF-?.Take tested ankle joint into formaldehyde for one week,then fixed it into EDTA for later use.After the joints are fixed,dehydrated and embedded it for paraffin section,then do the HE staining and immunohistochemical staining.Results:The circumference of rats' joints:It is knowed that,2 hours after the MSU modle completed,there are significant differences between the modle group and the blank group(P<0.01);24 hours after the MSU modle completed,there are significant differences between the high dose group of Tongfengxiao 2 and the modle group(P<0.01);48 hours after the MSU modle completed,there are significant differences between the high dose group of Tongfengxiao 2 and the modle group(P<0.01);72 hours after the MSU modle completed,there are significant differences between the high dose group of Tongfengxiao 2 and the modle group(P<0.01).The content of inflammatory mediator in rats' serum:compared the modle group with the blank group on content of IL-1 in rats' serum,there are significant differences(P<0.01);there are no differences between the high dose group of Tongfengxiao 2 and the modle group on content of IL-1 in rats'serum,but the value is lower;compared the modle group with the blank group on content of IL-6 in rats' serum,there are significant differences(P<0.01);there are significant differences on the content of IL-6 in rats' serum between the high dose group of Tongfengxiao 2 and the modle group(P<0.01);compared the modle group with the blank group on content of TNF-? in rats'serum,there are significant differences(P<0.01);there are also significant differences on the content of TNF-? in rats' serum between the high dose group of Tongfengxiao 2 and the modle group(P<0.01).The mean optical density of VCAM-1?ICAM-1 and NF-?B in rats' joints:compared the modle group with the blank group on mean optical density of VCAM-1?ICAM-1?NF-?B in rats' joints,there are significant differences(P<0.01);compared the high dose group of Tong Feng Xiao 2 with the modle group,the mean optical density of VCAM-1 and ICAM-1 in rats' joints are significant lower,and there are significant differences(P<0.01);compared the high dose group of Tongfengxiao 2 and the modle group,there are significant differences on the mean optical density of NF-?B in rats'joints(P<0.01).Pathological change of the rats' joints:observing under the light microscope,the high dose group of Tongfengxiao 2 showed that there are some inflammations in the jonts,and synovium of the joint is patially multiplication.Conclusion:1.The high dose group of Tongfengxiao 2 can induce the swell of the MSU modle rats' joints.2.The high dose group of Tongfengxiao 2 can suppress the the content of TNF-??IL-1? and IL-6 in the rats' serum.3.The high dose group of Tongfengxiao 2 can suppress the protein expression of VCAM-1?ICAM-1?NF-?B in the MSU modle rats' joints.4.Tongfengxiao 2 can suppress the inflammation in the MSU modle rats'joints and reduce hyperplasia in the synovium.5.The mechanism of Tongfengxiao 2 suppress the inflammation can be related to the signal path of NF-?B.Tongfengxiao 2 can suppress the produce of NF-?B,then restrain the produce of IL-1?,interfere with the inflammation cascade,and induce the releasing of inflammatory mediator such as TNF-? and IL-6,so it can suppresss the expression of ICAM-1 and VCAM-1.
