Purpose:To investigate the effect of the combined spinal cord acupuncture on the infiltration of macrophages and their expression of NgR in rats with spinal cord injury.Methods:The 72 Wister adult female rats were randomly divided into 4 groups:sham operation group,control group,electroacupuncture group,electroacupuncture+ running group.Each group consisted of 18 rats,and each group had three time periods.Each group had six rats.Only 10 segments of the thoracic spinal cord were exposed in the sham operation group,which was not broken,and the wound was sutured without treatment.The other three groups of rats in the chest 10 segments were all transverse to the spinal cord.The control group was not treated with any intervention.After the success of the model,the electroacupuncture group and the electric needle and the running table group were treated with the spine electroacupuncture treatment after the vital signs of the rats were stable,among which the electric needle and the running table group were given the training of the running platform in the second 2d after the molding.In 3 d,7 d,14 d after when using improved BBB movement function score evaluation rat hind limbs motor function recovery,then remove the damage of spinal cord tissue of rats each time point(6),and HE staining and SABC immunohistochemical staining method to detect and observe each macrophage infiltration,comparison between groups of macrophages expressing NgR.Results:1.Improved BBB score:?The function score of postoperative hind limb motor function was closely related to postoperative intervention,and BBB score was not significant before and after operation in the sham operation group.Compared with the control group,BBB score of the electroacupuncture group was significantly higher,and the difference was statistically significant(P<0.05).Compared with the electroacupuncture group,BBB scores of the electroacupuncture +running group were significantly higher,and the difference was statistically significant(P<0.05).?The functional score of postoperative hind limbs was closely related to the intervention time after surgery,and BBB scores were the lowest in each group,and the score improved at 7d and the highest score in 14d.2.HE staining:There were no infiltrated macrophages in the spinal cord of the sham operation group;In the spinal cord tissues of other experimental groups,infiltrated macrophages were present,and the changes of myelin were observed in the focal macrophages.After the damage of 3d,macrophages are mainly located in the area of injury,with less infiltration.After the damage of 7d,macrophages were concentrated in the area around the lesion,showing diffuse diffusion,and the cell volume was large,with a high density and zonal distribution,and the infiltration was significantly increased.14 d after injury,the macrophages mainly located around the damaged area and the central gray region and necrosis after the formation of the cavity wall,the regional distribution of cell volume smaller,there are still a lot of macrophage infiltration.3.SABC immunohistochemical staining method:?express NgR macrophages in the damaged area is closely associated with postoperative interventions,express NgR macrophages in the experimental group was significantly difference:the number of control is not found in the spinal cord tissue of express NgR macrophages.The number of macrophages in NgR was significantly increased compared with that in the control group,and the difference was statistically significant(P<0.05).Compared with the electroacupuncture group,the number of macrophages in NgR was significantly increased in rats in the electroacupuncture + rehabilitation training group,and the difference was statistically significant(P<0.05).?The expression of NgR was closely related to the time of postoperative intervention.In 3d,the number of macrophages expressing NgR was the least.At 7d,it gradually rises to the peak;At 14d,there was a downward trend.Conclusion:1.The combination of ridged electroacupuncture therapy and the combination of ridged electroacupuncture combined with treadmill training can improve the post-limb movement function of the injured rats after spinal cord injury.The combined treatment effect is superior to the chiropractic needle therapy.2.The treatment of clipping electroacupuncture and the combination of the clip with the spinal cord and the treadmill training can induce the expression of NgR in the area of the spinal cord injury in rats.The combined treatment effect is superior to the chiropractic needle therapy.
