Objective:To investigate the effects of Shen Fukang capsule on the renal interstitial fibrosis induced by unilateral ureteral obstruction(UUO),for providing theoretical treatment of chronic renal insufficiency in clinical.Methods:1.Established the unilateral ureteral obstruction model in rats:100 Male Sprague-Dawley(SD)rats were randomLy divided into sham operated control group,UUO model group,Shen Fukang capsule low dose group,Shen Fukang capsule middle dose group and Shen Fukang capsule high dose group;the rats in Shen Fukang capsule low,middle and high dose groups began to receive intragastric administration at day 7 after modeling.At days 14 and 21 after modeling,animals were sacrificed to determine the levels of scrum creatinine(Cr),blood urea nitrogen(BUN)and the N-acetyl-?-D-Glucosaminidase(NAG)in urine.HE-staining and Masson-staining were employed to observe renal pathologies changes.2.Impact of Shen Fukang on the renal interstitial fibrosis induced by unilateral ureteral obstruction:Immunnohistochemistry was used to detect Collenge ? and ?-SMA expression.3.Impact of Shen Fukang capsule on apoptosis of renal cell induced by unilateral ureteral obstruction: TUNEL staining was employed to detect the distribution of apoptotic cells.Western blot was used to detect Bcl-2 and Bax expression.And TGF-?1 and p38 MAPK expression were detected by immunnohistochemistry.Results:1.Established the unilateral ureteral obstruction model in rats:At days 14 and 21 after modeling,compared with the sham operated control group,the levels of scrum cr,BUN and urinary NAG of rat in the UUO model group were significantly increased(P<0.05),histopathology examination of renal tissue showed the renal injury was aggravated and the renal interstitial fibrosis was obvious,the renal tubulointerstitial injury index and the renal interstitial fibrosis area were obviously increased(P<0.05).Compared with the UUO model group,the levels of serum Cr,BUN and NAG in urine were significantly decreased(P<0.05)in Shen Fukang capsule low,middle and high dose groups,and the renal tubulointerstitial injury index and the renal interstitial fibrosis area were significantly decreased(P<0.05).2.Impact of Shen Fukang capsule on the renal interstitial fibrosis induced by unilateral ureteral obstruction:?-SMA and Collenge ? protein were mainly expressed in the cytoplasm of renal tubular epithelial cells.At days 14 and 21 after modeling,compared with the sham operated control group,the expression of?-SMA,Collenge ? protein of renal tissue were significantly increased in the UUO model group(P<0.05).Compared with UUO model group,the expression of ?-SMA,Collenge ? protein of renal tissue were significantly decreased in Shen Fukang capsule low,middle and high dose groups(P<0.05).3.Impact of Shen Fukang capsule on apoptosis of renal cell induced by unilateral ureteral obstruction: The positive cells of TUNEL staining,TGF-?1and p38 MAPK protein were all mainly expressed in the cytoplasm of renal tubular epithelial cells.At days 14 and 21 after modeling,compared with the sham operated control group,the apoptosis rate of renal cell was significantly increased(P<0.01)in the UUO model group,the expression of TGF-?1,p38 MAPK and Bax were significantly increased(P<0.01)while the expression of Bcl-2 was significantly decreased(P<0.05).Compared with the UUO model group,the expression of TGF-?1,p38 MAPK and Bax protein of renal tissue were significantly decreased in Shen Fukang capsule low,middle and high dose groups(P<0.05)while the expression of Bcl-2 was significantly incresed(P<0.05).Conclusion:1.Shen Fukang capsule significantly ameliorate against renal injury induced by unilateral ureteral obstruction,as evidenced by the decrease in the levels of scrum Cr,BUN and urinary NAG,as well as the attenuation of tubulointerstitial injury index and renal interstitial fibrosis area.The renal interstitial fibrosis induced by unilateral ureteral obstruction was made successfully.2.Shen Fukang capsule significantly reduce the renal interstitial fibrosis induced by unilateral ureteral obstruction,the possible mechanism may be inhibitting the deposition of ECM in renal tissue.3.Shen Fukang capsule can restrain renal cell apoptosis,which may play a protective role in reducing renal interstitial fibrosis induced by unilateral ureteral obstruction via inhibition of the TGF-?1/p38 MAPK signaling pathway.
