Objective Comparing the difference proteomics between adrenal cortical carcinoma and adrenocortical adenoma,normal adrenal tissue,to explore the pathogenesis of adrenal cortical carcinoma cancer,search for the biological markers associated with clinical diagnosis and the targets for treatment.Method Using solid phase PH gradient two-dimensional gel electrophoresis separation of adrenal cortical carcinoma and adrenocortical adenoma,normal adrenal tissue protein,after silver staining used ImageMaster 2DE software to find out differential protein spots.Then,differentially expressed protein spots were identified by mass spectrometry and compare with the Swissprot database.Results The results obtained by two-dimensional electrophoresis map has good repeatability.Adrenal cortical carcinoma tissue protein spots approximately 2196179,adrenocortical adenomaissue protein spots approximately2375 142 and normal adrenal cortical tissue protein spots approximately 2098152.There are 60 spots showed significantly different,of which 16 were Successfully verified.There are 6 protein up-expression in adrenal cortical carcinoma,include TUBB,PGAM1,CALB1,NNMT,HPT,CLUS.In contrary other are downexpression,PSB6,NDUS8,ST1A1,IF1AY,CYB5,CUTA,CATD,GSTO,GST2,IDHC.Compared with the adrenocortical adenoma,There are 35 protein show significant differences,11 protein were successfully identified,6 protein up-expression in adrenal cortical carcinoma,include HSPB1,PRDX6,NQ02,SOD,HNRH1,PGAM1.In contrary,others are down-expression,ANXA6,UCHL1,ATPA,ENOG-?,ST1A1.(P<0.01).Conclusion Differentially expressed proteins in adrenal cortical carcinoma aassociate with oxidation-reduction reactionredox,cell cycle and cancer-related genes.Tthese proteins are involved in tumor formation and invasion in adrenal cortical carcinoma.It may be the potential molecular markers for adrenocortical carcinoma diagnostic or therapeutic targets.
