Preliminary Functional Studies Of E2F1,RAGE And Its Downstream Effectors Expression In Diabetic Hepatocellular Carcinoma

Abstract/Summary:
Objective:The present study was designed to investigate the expression of E2F1,RAGE and its downstream oncogenes in hepatocellular carcinoma,and to explore the role of T2DM in the progress of HCC as well as its potential mechanism..Methods:Forty-eight hepatocellular carcinoma patients,who were confirmed by pathological examination after operation at the First Affiliated Hospital of Guangxi Medical University were included.The Patients were divided into two groups(Diabetic patients with Hepatic Carcinoma group(DM+HCC),n=18;non-diabetic group(HCC),n=30).The expression of RAGE,Rac1,AP-1,MMP-2 and MMP-9 in hepatocellular carcinoma and its paraneoplastic liver tissue were detected by immunohistochemistry.Furthermore,we explore the relationship between the differentiation and metastasis of HCC with these proteins.Western Blot was applied to analyze the expression of E2F1 protein in tissues of hepatocellular carcinoma and its paraneoplastic liver tissue.Results:(1)The level of RAGE,Rac1,AP-1,MMP-2 and MMP-9 proteins in cancer tissues of both DM+HCC and HCC groups were higher than those of their paraneoplastic liver tissues(P<0.05);Compared with the HCC group,RAGE,Rac1,AP-1,MMP-2 and MMP-9 expression in cancer tissues of the DM+HCC group were enhanced without significant difference(P>0.05).(2)The expression of RAGE,Rac1,AP-1,MMP-2 and MMP-9 in cancer tissues with metastasis were enhanced compared with those without(P<0.05).RAGE and Rac1 expression were not significantly different in differentiated cancer tissue.AP-1,MMP-2 and MMP-9 in poorly differentiated carcinoma were enhanced compared with those highly differentiated(P<0.05).Between two groups,the expression of RAGE,Rac1,AP-1,MMP-2 and MMP-9 were not significantly different whether there was metastasis or differentiation.(3)In both DM+HCC and HCC group,the relative expression of E2F1 in hepatocellular carcinoma tissue was higher than that in pericarcinomatous tissue;And compared with the HCC group,E2F1 expression was higher than that in DM+HCC group(P<0.05).Conclusions:1.There was no significant difference in the expression of RAGE,Rac1,AP-1,MMP-2 and MMP-9 between DM+HCC and HCC group,the signs showing whether there was metastasis or differentiation between two groups were also not different.It showed that RAGE might not be the main mechanism by which hyperglycemia increase the risk of HCC in diabetes.2.Both in DM+HCC and HCC group,the expression of E2F1 was significantly increased in cancer tissues.Compared with the HCC group,E2F1 expression in cancer tissues of the DM+HCC group were enhanced.E2F1 may play a role in the progress of HCC patients with DM.
