Objective:To explore the clinical diagnosis, targeted therapy and prognosis evaluation of INPP4 B in EOC,we examed the expression of INPP4 B in different groups, and analysed the significance between the expression of INPP4 B and clinical parameters.Method:A retrospective analysis was made of 182 cases of epithelial ovarian cancer treated from January 1st 2006 to December 31 st 2013 and 20 cases of benign ovarian tissue treated with other diseases at the same time.Immunohistochemical method was used to detect the expression of INPP4 B in various epithelial ovarian cancer tissues and to compare the relationship between the expression of INPP4 B and clinical parameters,to compare the relationship between INPP4 B expression and clinical classification, clinical stage, histological classification, preoperative CA125 level, preoperative CA125 mean, postoperative recurrence, survival status, and sensitivity of platinum chemotherapy in patients with epithelial ovarian cancer. Results: There were 182 epithelial ovarian cancer cases, including 121 cases of serous carcinoma(66.5%),30 cases of mucinous carcinoma(16.5%),16 cases of endometrial carcinoma(8.8%),and 15 cases of clear cell carcinoma(8.2%).There were 83 cases of type I ovarian cancer(45.6%),including 25 cases of low-grade serous carcinoma,30 cases of mucinous carcinoma,13 cases of low-grade endometrial carcinoma, and 15 cases of clear cell carcinoma. There were 99 cases of type II ovarian cancer(54.4%),including 96 cases of high-grade serous carcinoma and 3 cases of high-grade endometrial carcinoma. The average age of type I was 56.0,and type II was 59.9,p=0.017.In type I,there were 55 patients(66.3%)in early clinical stage and 28 cases(33.7%)in late stage. In type II,there were 28 cases(28.3%)in early clinical stage and71 cases(71.7%)in late stage,p=0.000.In type I,there were 68 cases(81.9%)with preoperative serum CA125 <500U/ml,and 15 cases(18.7%)CA125≥500U/ml;there were 48 cases(48.5%)CA125 <500U/ml,51 cases(51.5%)CA125≥500U/ml in thetype II,p=0.000.The average survival time of type I was 63.3 months, versus type II was48.8 months,p=0.000.Immunohistochemical staining showed that INPP4 B was expressed in cytoplasm. The high expression rate of INPP4 B in 20 normal ovarian tissues was 60.0%and only 36.8% in 182 cases of epithelial ovarian cancer. There was statistical significance between two groups(p=0.044).There were 41 cases(49.4%)with no or low expression of INPP4 B in type I epithelial ovarian carcinoma group,42 cases(50.6%)with high expression in type II group and 74 cases(74.7%)with no or low expression in type II group. The expression rate of INPP4 B in type II group was significantly lower than that in type I group(n = 25).The expression rate of INPP4 B in type II group was lower than that in type I group(p=0.000).Further analysis of the expression of INPP4 B in different histological types revealed that, the percentage of high expression in 121 serous carcinoma(31.4%),30 mucinous carcinoma(60%),16 endometrial carcinoma(37.5%)and 15 clear cell carcinoma(33.3%).The expression of INPP4 B in mucinous ovarian cancer was significantly increased, which was not consistent with the other three tissue types(p=0.036).The median survival time was 57.0 months in no or low expression of INPP4 B,and 104.0 months in the high expression group. In addition, there was obvious difference between the two groups in the 5 year survival rate, which was 35.4% in no or low expression of INPP4 B,and 62.0% in the high expression group. There was no correlation between expression of INPP4 B and FIGO stage、 recurrence、platinum resistance. After exclusion of mucinous carcinoma, the expression of INPP4 B was correlated with the classification of "dualistic model"(p=0.012)、5-year survival rate(p=0.001).The total survival status of the high expression group was better than that of in no or low expression group, and there was associated with the FIGO stage 、preoperative CA125 level、recurrece time but no correlation in platinum resistance. Conclusions: 1.The expression of INPP4 B in cytoplasm was detected by immunohistochemistry.Compared with normal ovarian tissues, the expression of INPP4 B in epithelial ovarian carcinoma was lower than that in normal ovarian tissues. The expression of INPP4 B in epithelial ovarian carcinoma, including serous 、endometrial and clear cell carcinoma, was mostly no or low expressed, but only mucinous carcinoma showed relatively high expression.2.There was a significant correlation between the expression of INPP4 B and the classification of "dualistic model" in epithelial ovarian carcinoma, and the 5 year survival rate of patients with no or low expression of INPP4 B was lower than that of type I.The results suggest that the expression of% INPP4 B is related to the prognosis of epithelial ovarian cancer.
