The Study About The Protective Effect Of San-Han Huo-Xue Qu-Bai Recipe And Its Ingredients To The A375 And PIG1 Cells Induced By Hydrogen Peroxide Against Oxidative Damage

Objective To explore the protective effect of San-Han Huo-Xue Qu-Bai Recipe and its ingredients to the melanocyte injury under oxidative stress.Methods Different concentrations of H₂O₂?100?M,200?M,500?M and1000?M?were added to Melanocytes and acting on them for 24h,36h and72h respectively,then the optimum concentration and time of oxidative damage was determined,and an oxidative stress model was established for the following.The experiment was divided into blank control group,H₂O₂ model group,San-Han Huo-Xue Qu-Bai Recipe group and its ingredients group?Include Yang-Xue Huo-Xue group,San-Han Wen-Yang group,Bu-Yi Gan-Shen group?of different concentration?1mg/mL,2mg/mL,4 mg/mL,8mg/mL,16 mg/mL and 32 mg/mL?and vitiligo capsule group.CCK8 was used to detect the protective effect of drugs from different groups on cell damage induced by H₂O₂.Results All of the San-Han Huo-Xue Qu-Bai Recipe and its ingredients can effectively reduce the oxidative damage on A375 cells from H₂O₂,And the protective effect was most obvious when the concentration was 32mg/ml.Under the same concentration,the protective effect of the San-Han Huo-Xue Qu-Bai Recipe group was similar to that of the vitiligo capsule group,and its ingredients group were weaker than the vitiligo capsule group?P<0.05?.The protective effect from all of groups for PIG1 cells were weaker than for A375 cells.And there wsa no obvious protective effect from the San-Han Wen-Yang group and Bu-Yi Gan-Shen group.San-Han Huo-Xue Qu-Bai Recipe group and all of its ingredients group were weaker than vitiligo capsule group.ConclusionSan-Han Huo-Xue Qu-Bai Recipe group and all of its ingredients group can significantly inhibited the A375 cell damag induced by H₂O₂.And the protective effect of the San-Han Huo-Xue Qu-Bai Recipe group and vitiligo capsule group was similar,A few other groups were slightly weaker.The protective effect of San-Han Huo-Xue Qu-Bai Recipe group and all of its ingredients to the PIG1 cells againstoxidative damage loaded by H₂O₂ is weaker than that of the vitiligo capsule group.
