Objective The IL-11/STAT3 signal transduction pathway has been reported to play an important role in human gastric cancer,and STAT3 regulates mitochondrial activity either positively or negatively in cancer cells.In this study,the expression of IL-11,STAT3 and p-STAT3 in gastric mucosa of chronic superficial gastritis,mild to moderate dysplasia,severe dysplasia of gastric mucosa epithelium and gastric cancer were detected by transmission electron microscopy Gastric mucosal epithelial cells mitochondrial ultrastructure changes,to further explore the changes of IL-11/STAT3 signaling pathway and mitochondrial ultrastructure in human gastric mucosa epithelial dysplasia and gastric cancer.Methods We collected gastroscopy and pathological biopsy diagnosed as chronic superficial gastritis,mild to moderate dysplasia of gastric mucosa epithelium,severe dysplasia of gastric mucosa epithelium and gastric mucosa in gastric antrum or gastric mucosa in 60 patients,divided into 4 groups,15 cases in each group.The expressions of IL-11,STAT3 and p-STAT3 in gastric mucosa of 4 groups were detected by immunohistochemical method.The changes of mitochondrial ultrastructure in gastric mucosal epithelium of some groups were observed by transmission electron microscope.Results(1)Immunohistochemical analysis showed that the positive rates of IL-11 in gastric dysplasia group(80.00%,12/15)and GC group(86.67%,13/15)were significantly higher than those in CSG group(33.33 %,5/15),p <0.05,while in gastric mucosa epithelial mild to moderate dysplasia group(60.00%,9/15)and CSG group as well as mild to moderate dysplasia group,severe dysplasia group,GC group(P> 0.05).The statistical analysis also showed that the high expression of IL-11 was associated with Hp infection(P <0.01),but not with the gender,age(P> 0.05).The expression of STAT3 in gastric mucosal epithelial dysplasia group(80.00%,12/15),gastric mucosa epithelial dysplasia group(86.67%,13/15)and GC group(93.33%,14/15)were significantly higher than those in CSG group(33.3%,5/15),p <0.05,while those in mild to moderate dysplasia group,There was no significant difference in the positive expression rate of STAT3 between the severe dysplasia group and the GC group(p> 0.05).The high expression of STAT3 and the combination of H(P <0.05),but had no correlation with the gender,age,differentiation of gastric cancer and lymph node metastasis(p> 0.05).The expression of p-STAT3 in CSG group The positive rate of p-STAT3 expression in CSG group was significantly lower than that in GC group(80.00%,12/14),but lower than that in severe dysplasia group(73.33%,11/15)/15),p<0.01.The positive rate of p-STAT3 in mild to moderate dysplasia group was not significantly different from that in CSG group(p> 0.05).The expression of p-STAT3 in mild to moderate dysplasia group,severe dysplasia group and GC group The positive rate of p-STAT3 was not significantly different(p> 0.05).The high expression of p-STAT3 was associated with Hp infection(P <0.01),but not with the gender,age,And lymph node metastasis no significant correlation(p> 0.05).(2)The number of mitochondria in the gastric mucosal epithelial cells decreased,swelling,vacuolar degeneration,matrix fades,pyknosis,mitochondrial cristae rupture and disordered degeneration were observed under transmission electron microscope.The degree of degeneration was higher in CSG and gastric mucosal epithelium Mild to moderate dysplasia,severe dysplasia of gastric mucosa epithelium and GC gradually increased,the statistical analysis results suggest that gastric mucosal epithelial dysplasia group,gastric mucosa epithelial dysplasia group and GC group of gastric mucosa mitochondria The incidence of degeneration and number of vacuolar degeneration was significantly different from that of CSG group(P <0.05).Conclusion The IL-11/STAT3 pathway may play an important role in the occurrence and progression of severe gastric dysplasia and gastric cancer.It may play a role in the prevention and treatment of gastric mucosa by regulating mitochondrial activity resulting in the degeneration of mitochondria.Dysplasia and gastric cancer provide a new target,has important clinical application prospects.
