Objective1-bromopropane(1-BP)is similar to freon,and has the advantages of low boiling point,volatile,stable chemical properties,and not easy to burn.At the same time,it does not destroy the atmospheric ozone layer.EPA has identified it as one of the alternatives to freon,which has been rapidly promoted and the use has increased year by year.Toxicological studies have shown that 1-BP has neurological and reproductive toxicity.A number of cases of severe neurological impairment in occupational population exposed to high concentrations of 1-BP had been reported both at home and abroad.China is a big country in the produce and use of 1-BP.There is a large number of occupational exposure groups.1-BP exposure has become a health risk factor of occupational population.The occupational exposure limits,biological contact limits and corresponding detection methods of 1-BP in the air of workplaces have not been formulated in china.This study evaluated the feasibility of 3 biomarkers(1-BP,Br,AcPrCys)as biological monitoring indicators of occupational exposure to 1-BP,to establish a reliable marker as a biological monitoring indicator for occupational exposure to 1-BP.MethodsGuidelines for the determination of chemical substances in biological materials in accordance with the guidelines for the establishment of GBZ/T210.5-2008 occupational health standards(fifth part),the linear range,detection limit,precision,accuracy,interference and stability,verification and field application tests of three biomarker detection methods were carried out respectively,the indexes of 3 biomarker detection methods were optimized and established.66 people exposed to 1-BP were used as exposure group,and 66 non 1-BP field workers served as control group.The concentrations of three biomarkers in the urine samples of control group and contact group were detected,and the concentration of 1-BP in the air of the exposed group was also measured.The levels of three biomarkers in the control group and the contact group were analyzed.The correlation between the concentrations of biomarkers and the concentration of 1-BP in the air of workplace was analyzed.Finally,reliable biological monitoring indicators of occupational exposure to 1-BP were established.ResultsThe concentration of 1-BP in urine was linear in the concentration range of 50?1000?g/L,the correlation coefficient was 0.9992,the detection limit was 5?g/L,the recovery rate was 92.4%?104%,the intra-batch precision was 4.5%-8.7%,and the inter-batch precision was 5.3%?9.8%.The linear range of AcPrCys in urine was 15?1000?g/L,the correlation coefficient was 0.9996,the detection limit was 5pg/L,the recovery rate was 88.6%?92.6%,the intra-batch precision was 2.7%?6.2%,and the inter-batch precision was 4.5%-8.1%.The linear range of bromide ion in urine was 15?1000?g/L,the correlation coefficient was 0.9992,the detection limit was 2?g/L,the recovery rate was 95.6%?98%,the intra-batch precision was 1.4%?3.2%,and the inter-batch precision was 3.4%?5%.The three kinds of biomarker detection methods are in line with the requirements of GBZ/T210.5-2008 guidelines for occupational health standards fifth part:determination methods of chemical substances in biological materials.There were significant differences in the concentrations of three biomarkers between the control group and the contact group(P<0.05).The concentrations of 1-BP and AcPrCys in urine were significantly correlated with the concentrations of 1-BP in air.The concentration of Br-in urine is not correlated with the concentration of 1-BP in air.The instruments needed for urine AcPrCys detection are expensive and difficult to popularize.The background value of bromine ion in the urine of the control group was higher.ConclusionUrinary 1-BP can be used as reliable biological monitoring indicator for occupational exposure to 1-BP.Urinary bromine and AcPrCys can be used as reference indicators for occupational exposure to 1-BP.
