Background HCC is one of the common malignant tumors in the digestive system and it has the poor prognosis.At present,there has been no definite standards and guides evaluating prognosis at home and abroad.The predicting system for HCC prognosis plays an essential role in the decision of individualized treatment scheme.FGF 19 is a peptide molecule and plays an important role on tumor generation and development.Ultrasound contrast has the function of excellent blood flow development.By combining with quantitative analysis of ultrasound contrast,time-strength curve is obtained to quantitatively embody blood flow situation of focus vessel.The research aims to discuss the clinical value of FGF19 in HCC and discuss the value of using quantitative analysis of ultrasound contrast to evaluate HCC prognosis.Method 1.Correlation between quantitative analysis of ultrasound contrastand FGF19.1.1 The ultrasonic-contrast image of 30 HCC patients were summarized,IMAX,mTT,TTP,RT,RS were accqured by TIC(Time-intensity curve),with the combinetion with conduct quantitative analysis,can reflect the blood flow of the lession.1.2 Tissue microarray was used to detect immunohistochemistry andpostoperative specimens of 30 patients included in this study were collected.The expression of FGF19 protein in HCC tissues and normal liver tissueswas detected by immunohistochemistry.1.3 Differential expression of FGF19 protein between HCC and normal livertissues was analyzed by chi-square test.Fisher exact propability was used toanalyze the correlation between quantitative analysis of ultrasound contrastand the expression of FGF19.2 Bioinformatics2.1 FGF19 gene expression profile and clinical parameter information was downloaded from TCGA database2.2 Clinical significance of FGF19 in TCGA databaseGEPIA was used to compare with the FGF19 expression level between HCC group and normal liver tissue group.Human protein atlas website was used for survival analysis of FGF19 to generate the survival graph.Chi-square test was used to detect FGF19 expression law and correlation with clinical pathological parameters.Clinical value of FGF19 expression level taken the median,P<0.05,showing that there is the statistical difference.2.3 Prediction of FGF19 relevant gene,gene enrichment analysis and functional annotationMEM website was used to search for Related genes and conduct GO analysis and KEGG enrichment path analysis in the DAVID database.2.4 Gene mutation level detection and regulation network constructionGenomics website cBioPortal was used to analyze FGF19 changes of mutation and copy number in FGF19 and construct the regulatory network for relevant genes of FGF19.Resulted There was significant difference in the expression of FGF19 protein between HCC and normal liver tissues(P<0.05).FGF19 was over-expressed in HCC.FGF19 positive expression group was related to RS,TTP and RT in quantitative analysis of ultrasound contrast.There was the statistical significance(P<0.05),but it was irrelevant of IMAX and mTT.The statistical results of TCGA database showed that FGF19 expression in HCC tissues was present in the high-expression tendency.There was no significant difference in the high expression of FGF19 with age,gender,tumor TNM stage,and tumor cell grading(P>0.05).There was no statistical significance between total survival time of HCC patients with FGF19 high expression and with the low one,but the former was shorter than the latter.Co-expression genes obtained from MEM website conduct GO annotation and KEGG enrichment analysis.Related genes participate in DNA transcription,phosphatidylinositol-4,5,5,ethyl phosphoric acid kinase activity and Golgicomplex(P<0.02).KEGG path analysis was enriched cancer path.Genomic website analysis was used to analyze variation situations of FGF19co-expression gene in HCC.The results showed that 12% of FGF19 taken place gene mutation to participate in mitosis of cells and it was present in negative correlation with DNA methylation.By constructing the regulatory network,it could be found that variation frequency of PTK2,PIP5K1 A and SDC2 was relatively high.Conclusion:The expression of FGF19 was highly expressed in HCC.It may be involved in the occurrence and development of HCC through relevant target genes,suggesting that FGF19 may be a potential adverse prognostic risk factor;Through the combination of quantitative analysis of contrast-enhanced ultrasound parameters and FGF19 expression,to understand the prognosis of HCC patients,provides a basis for individual treatment of HCC.
