| In recent years,metabonomics,which has been an important part in life science and developing gradually supported by the increasingly innovative analysis techniques,is becoming an significant and valuable tool in biological research.Meanwhile,the research on the clinical application of bee venom is rising;as a kind of substance provided with highly pharmacological and biological activities,bee venom has significant curative effects on rheumatism,and also shown a great potential in the clinical treatment of cancer.Nevertheless,there is still a larger development space for the studies on the bee sting as well as the effect of bee venom constituents on the endogenous metabolism of living organism.To explore the influence of bee venom to endogenous metabolism,this research used liquid chromatography-mass spectrometry to analyze the changes of metabolites and possibly involved metabolism pathway in rats serum.The serum samples of SD rats in the blank group,the trauma group and the experimental group were analyzed,and the relevant difference metabolites were selected by means of ROC value test and T-test.The hormesis of bee sting to an organism can be roughly divided into two aspects,namely,the hormesis of bee venom constituents and sting injury,this research studied the effect of bee sting on the endogenous metabolism in rats by taking bee venom constituents and sting injury as two variables from the perspective of metabonomics,and determined that,compared with sting injury,bee venom constituents are the main factor influencing the endogenous metabolism in rats in the complete bee sting process.It also probed into the metabolism in stung rats,and found the symbolic discrepant metabolites and pathways related to the constituents of bee venom.After modeling and statistical analysis with PLS-DA under positive and negative ions models,it was discovered that there was unexceptionally significant difference in metabolite for the Sting Group,Traumatic Group and Blank Control Group.It can be concluded that bee venom components,sting behaviors and sting trauma exert unexceptionally a significant disturbing action on mice endogenous metabolites.In-pair differential metabolite screening was conducted respectively among the three groups of serum samples.To probe into the potential biomarkers related to bee venom components,differential metabolite screening was conducted to the Sting Group and the Traumatic Group.It was discovered that there were 327 kinds of screened differential metabolites under positive and negative ions models.After structural identification and further screening to such differential metabolites,totally 39 differential metabolites were believed to be associated with bee venom component stimulation.To probe into the potential biomarkers related to complete bee-sting process,differential metabolite screening was conducted to the Sting Group and the Blank Control Group.It was discovered that there were 164 kinds of screened differential metabolites under positive and negative ions models.After structural identification and further screening to such differential metabolites,totally 71 differential metabolites were believed to be associated with complete bee-sting process.To probe into the potential biomarkers related to Traumatic,differential metabolite screening wasconducted to the Sting Group and the Traumatic Group.It was discovered that there were 107 kinds of screened differential metabolites under positive and negative ions models.After structural identification and further screening to such differential metabolites,totally 48 differential metabolites were believed to be associated with complete bee-sting process.After the process of further screening,nine potential biomarkers which show relation with bee venom were found,including carnitine,acetylcarnitine,lPC,LPA,lyso-PC,lyso-PE,IL-20,linoleic,sphinganine,and four mentabolic pathways were discovered,which are linoleic acid metabolism,sphingolipids catabolism,phospholipid catabolism,alanine metabolism and aspartate metabolism.The research indicates that sting,stabbing and bee venom all have influence on endogenous metabolites of SD rats and bee venom,comparing with stabbing,shows stronger effect.Bee venom plays an important role in the metabolic process of fatty-acid and endogenous metabolism will also be influenced through this process.Therefore,according to physiological function of affected metabolism pathway,exploration about this process has significant meaning to physiology. |
