AIM:Objective to investigate the expression of a-defensin-2 mRNA(rBD2)in the lung tissues and the changes of interleukin 2(IL-2)in peripheral blood and the content of SP-A and sIgA in bronchoalveolar lavage fluid(BALF)in rats with tracheal intubation and the intervention effect of Epimedium Polysaccharide.METHODS: The healthy male Sprague-Dawley(SD)SPF rats of 54,were fall into 3 group: the normal group(A),the tracheal intubation group(B)and the tracheal intubation group(C),A,B,C group(n = 18).To establish a rat model of tracheal intubation.After successful modeling,the 24 h,72h,168 h after drug delivery to get the sample.The peripheral blood samples,lung tissue and bronchoalveolar lavage fluid were collected to detect the expression of rat ?-defensin-2 mRNA(rBD2)in the lung tissues,interleukin2(IL-2)in peripheral blood of rat,SP-A and s IgA in the lung lavage fluid of the lung tissues of rats.RESULTS:(1)in the group A,the result of lung tissue biopsy were normal;in the group B and C,all have different degrees of edema,alveolar and interstitial inflammation,hemorrhage,pulmonary atelectasis and pulmonary tissue injury atthree phases(24h,72 h,168h),group B showed pulmonary interstitial and intra alveolar hemorrhage,inflammatory cell infiltration,alveolar septum etc.As time goes by,the damage is increasing.In group C,lung injury such as alveolar hemorrhage or destruction,inflammatory cell infiltration could be seen,but the pathological changes of 72 h and 168 h in two groups were lighter than those in group B.(2)Convey of rBD2 in lung tissue of rats was obviously imporved at24 h phase after tracheal intubation,and then decreased,especially after 72 h,and decreased to the lowest value at 168 h.The expression of 24 h was significantly increased after drug administration,and the expression reached the highest value after 72 h,but decreased after 168 h,but was still higher than that in the non drug group and the normal group.(3)The content of IL-2 in serum of rats was increased after first days after tracheotomy,and then no change,but the content increased significantly with the time.(4)The concentration of SIgA in BALF in rats with concentration in tracheotomy catheter began to increase at the first24 h,then reached the peak at 72 h and decreased to the minimum value after168 h,which is lower than the level of normal control group;group C after medication with its expression in the 24 h began to increase,and increased with the prolongation of time.(5)The concentration of SP-A in BALF in rats with the tracheotomy catheter increased significantly,with the extension of tracheotomy intubation time decreased(P < 0.05),group C after the drug intervention,the expression of SP-A in the 24 h began to rise,reached the highest value in the 72 h,in the 168 h decreased but was still higher than that of group A and group B(P <0.05).CONCLUSION: Effect of Epimedium Polysaccharide on immune function of rats after tracheotomy,especially the local immune function of lung.The immune enhancement of Epimedium Polysaccharide may be associated with thetracheotomy catheter expression in lung tissue of rats with beta defensin-2mRNA,the improvement of the content of sIgA in BALF and interleukin 2(IL-2)in peripheral blood,and resistance in bronchoalveolar lavage fluid of SP-A degradation.
