Objective:In this experiment,male KM mice were used as experimental subjects to understand the effects of treadmill movement and flavonoids from blackberry wolfberry on brain tissue of depressed mice model,and the Toll-like receptor pathway in the immune system The Mechanism of Factor.Methods:In this study,the mouse model of depression was modified by the Wilier CUMS model.Sugar consumption,open box experiment,forced swimming,hanging tail for model validation.The mice were randomly divided into two groups:blank group(OG),model group(M),treadmill group(TM),blackberry wolfberry flavonoids group(LRMF),treadmill combined with blackberry wolfberry flavonoids group(LRMF+ TM),each group of 10.Mice were examined and recorded before and after intervention,and the changes of diet,activity and nerve excitability were observed.The mice were fasted overnight.The contents of TLR4 and 5-HT in the blood were measured by ELISA.Pathological sections were used to observe the distribution of Nissl bodies in the brain tissue.Immunohistochemistry was used to detect the expression of MyD88 and TNF-a in Toll-like receptor signaling pathway IL-10,and the expression of MyD88,TNF-ca and IL-10 in brain tissue were detected by Realtime-PCR.To investigate the protective effects of treadmill and blackberry wolfberry flavonoids on the brain tissue of depressed mice and the influence of Toll-like receptor signaling on the immune system.Results:? Behavioral test:Compared with oG group,the consumption of sugar in each group decreased,the swimming time in forced suspension,hanging tail and open box experiment increased significantly and the excitability decreased(P<0.01)(P<0.05),followed by TM group,LRMF group and M group(P<0.05),and the difference was significant(P>0.05)·? ELISA test results:The level of rLR4 in LRMF + TM group was significantly lower than that in M group(P<0.05).The level of LRRF + TM group was significantly lower than that of M group(P<0.05)The levels of 5-HT in TM group were the highest(P<0.05),followed by TM group and LRMF group(P<0.05).? Nissl staining results showed:OG group in the number of Nissl body more and the location of normal,nuclear-free shrink,disappear phenomenon.(P<0.05).Compared with the M group,the number of LRMF + TM group was significantly lower than that of the M group(P<0.05).The LRMF +TM group had significant nuclear fixation(P<0.05),and the number of them was significantly lower than that of the control group(P<0.05).?immunohistochemical results:The expression of MyD88 and TNF-a protein in the brain tissue of LRMF + TM group was the lowest(P<0.05),LRMF group and TM group were lower than those of M group(P<0.05)The expression of IL-10 protein was higher than that of M group,and the expression of IL-10 was the highest(P<0.05).?Real-time PCR results:The expression of MyD88 and TNF-a mRNA in LRMF + TM group,LRMF group and TM group was lower than that in M group(P<0.05),and the expression of IL-10mRNA in LRMF +(P<0.05),followed by OG group>LRMF + TM group>TM group>LRMF group>M group.Conclusion:Treadmill movement and herbal flavonoids have protective effects on the brain tissue of depressed mice,which can enhance the excitability of the brain and the immune function of the brain.The mechanism of action may be effective in inhibiting the expression of MyD88,Thus promoting the relevant immune factors,brain neurons protective factor expression,reduce depression symptoms.
