Objectives The exeistence of intestinal ganglion cells is an important basis for diagnosis of Hirschsprung'S Disease. It is also important to guide surgical excision of intestinal tract. This text makes use of diffrent neural ma kers to study the distribution of SY?S-100?Nue-n in the control group and t he ganglionic bowel segment and aganglionic segment of HD group, then anal yzes and concludes the advantages and disadvantages of diffrent immunohistoc hemical method, exploring the diagnoseic value of Nue-N in Hirschsprung'S Disease, and make it better to be used in diagnosing and guiding treatment. Methods The colon speciments of 30 csaes of HD and 2 cases of accidental d eath without digestive tract malformations were collected. Then use the method of immunohistochemical to detect the expression of SY?S-100?Nue-n in the tiss ue of the control group and the ganglionic bowel segment and aganglionic segm ent of HD group. At the last, analyse the statistical data. Results 1.The expansion segment of HD lack ganglion cell, the nerve giber gets thicker than the other group.Form the narrow segment to the expansion segment, the number of ganglion cell increases.2.There were statistical differences in the expression of three kinds of imm unological markers in the congenital megacolon and the control group.3.Nue-n only can express in the nucleus of ganglion cell,on the contrary,S-100 cannot express in the ganglion cell, but it can distribute in the nerve giber a nd gliocytes. The cytolymph of ganglion cell can be expressed with SY.4.The expression of Nue-N in the narrow segment of the congenital megacolon and the transfer segment was statistically different from the control group,a nd the difference between the expansion segment and the control group was not obvious.5.There was no statistical difference between Nue-N and SY immunohistoch emistry in diagnosis of congenital megacolon stenosis. Conclusions 1.The expression distribution of Nue-N in the intestinal segments o f congenital megacolon was consistent with the distribution of ganglion cells in t he congenital megacolon.2.Nue-N immunohistochemical staining can specificity shows ganglion cells form, more intuitive than other immunohistochemical staining methods, the diag nosis and differential hirschsprung homogenous provides new method worthy of f urther study.3.The combination of different immunohistochemical methods can more acc urately diagnose congenital megacolon.4.SY,Nue,S-100-N in the expression of congenital megacolon is unus ual, speculated that the pathogenesis of congenital megacolon and enteric nervous system development in the process of SY,Nue,S-100-N related expressed genes are closely related.
