Differences In Expression Of Fibulin-5 Protein In Pelvic Organ Prolapse In Xinjiang Uyghur And Han Women

Objective:1.This study intends to determine fibulin-5 which express in connective tissue of vaginal anterior wall the Uyghur and Han POP patients,to discuss the role of occurrence and development of pelvic support tissue structural and metabolic changes in POP.2.To analysis and make clear whether the expression of fibulin-5 has difference between the two populations,Analysis and summarizes the characteristics of the protein in gene expression and clinical significance.This study means a lot to clarify the POP etiology and racial diversity research,and plays a preventive role in etiology.Materials and Methods:1.Materials: Collected the pelvic organ prolapse patient who has treated in the First Affiliated Hospital of Xinjiang Medical University,gynecology 72 patients from July 2013 to December 2015 as Experimental group;128 cases of hysterectomy due to benign lesions of the uterus were taken as control group.2.Methods:2.1 Using immunohistochemical method,the expression of fibulin-5 in the protein level of the anterior wall of the vagina.2.2 Reverse transcription polymerase chain reaction(RT-PCR)method was used to detect the expression of fibulin-5 in the gene level.Results:1.Immunohistochemical method :The fibulin-5 were expressed in POP patients and non POP patients with anterior vaginal wall tissue,but the expression of the experimental group was significantly less than that of the control group(p <0.05),while comparing the different nation of experimental group and control group respectively,the difference of fibulin-5 expression had no statistically significant(p>0.05).2.RT-PCR: fibulin-5 mRNA expression was positive in the four groups.In the experimental group and the control group,the Uyghur experimental group and the Han experimental group,the Uyghur control group and the Han control group,the difference was statistically significant(p<0.05).Conclusion:1.The expression of the fibulin-5 protein in the experimental group and the control group has statistical significance,indicating that fibulin-5 protein plays an important role in maintain supporting tissue of pelvic cavity functions,2.Between the Uyghur and Han in the experimental group and the control group respectively difference were not statistically significant in immunohistochemical method,but were statisticallysignificant in RT-PCR,indicating that the relevant factor in the pathogenesis of pelvic organ prolapse disease might have racial differences,to provide support and assistance to the research on the etiology of pelvic floor dysfunction disease.
