| Objective: We established a radioresistant subclone cell line(SCC10a-IR)derived from laryngeal cancer cell line SCC10 a by treating the cells with five rounds of sublethal irradiation.Using the clone survival test and cell proliferation assay to determine the radiosensitivity of SCC10 a cells and SCC10a-IR cells,and to compare the differences in invasion and migration of SCC10 a cells and SCC10a-IR cells.In order to clarify the mechanism of radioresistance in laryngeal cancer.Methods:After confirming the sublethal irradiation dose of SCC10 a cells,SCC10a-IR cells were constructed by treating with five rounds of sublethal irradiation.Using the clone survival test and cell proliferation assay to compare survival rate of SCC10 a cells(control group)and SCC10a-IR cells(experimental group),and clarifing the changes of invasion and migration by using the scratch test,transwell invasion and migration experiments.Results: 1.Established a radioresistant subclone cell line-SCC10a-IR,Which derived from laryngeal cancer cell line SCC10 a by treating the cells with five rounds of sublethal irradiation.The results of clone survival test and cell proliferation assay showed that the radiation resistance of SCC10a-IR cells was significantly higher than SCC10 a cells.2.The change of invasion and migration in SCC10 a cells and SCC10a-IR cells.The scratch test,transwell invasion and migration experiments showed that the longitudinal migration ability,lateral migration ability and invasion ability of SCC10a-IR cells(experimental group)were significantly higher than those of SCC10 a cells(control group).Conclusion:1.Established a radioresistant subclone cell line-SCC10a-IR.2.SCC10a-IR cells were significantly higher than SCC10 a cells in migration and invasion ability. |
