| Backgrounds and Objective: The proinflammatory monocytes also names Fc?RIII,there are related research prove in bacterial sepsis,tuberculosis,dengue fever,around the retina vein inflammation,abnormal expression in diseases such as rheumatoid arthritis.the CD14 +CD16 + were labeled proinflammatory based on higher expression of proinflammatory cytokines and higher potency in antigen presentation.There are many cells who express different types or kinds of Ig Fc receptors,and they have very important roles in promoting and regulating immune.The positive feedback regulation of many immunocytes are promoted by the activation of proinflammatory monocytes,though immunoreceptor tyrosine-based activation motif(ITAM)or not,it can promote the activation of activating Fc? receptor.More and more studies have shown that in the pathogenesis of tumor and autoimmune diseases(such as rheumatoid arthritis,systemic lupus erythematosus,sjogren syndrome),the proinflammatory monocytes plays very important role.But several studies have also indicated that inflammatory mononuclear cells associated in induction of inflammation and immune complex the release of inflammatory factors play an important role.With lupus nephritis(LN)is a systemic lupus erythematosus(SLE)of kidney damage,the main pathogenesis of formation and deposition of immune complex(IC),so we consider whether inflammatory mononuclear also involved in the pathogenesis of LN.To open the possible pathogenesis of LN,our research find the different expression of proinflammatory monocytes in LN and HC,we also find the correlation with proinflammatory monocytes and patients' disease activity.To find mononuclear inflammatory on the pathogenesis of LN,the possible molecular mechanisms,we detected the expression of proinflammatory monocytes after treatment of six months.To clarify the relationship between proinflammatory monocytes and LN will also give new area to the new treatment methods for LN.The results could get we more clear to the pathogenesis of LN and provide new targets for therapy.Contents and Methods:We recruited LN patients who have not get any therapy,and the age and sex matched to healthy controls(HC).We use the Flow cytometry to detecting the expression of proinflammatory monocytes on the monocytes from the whole blood of HC and LN.The correlation between the expression of proinflammatory monocytes with the disease activity indexes of LN were analyzed.Monocyte subpopulations were sorted to detect the level of inflammatory cytokines.For further study,after treatment of six months.,we detected the proinflammatory monocytes' expression on the monocytes.Results: In LN patients,The expression of proinflammatory monocytes was significantly higher than HC(MFI 1970±835 vs 1320±252,p=0.0085);It was negatively with the level the expression of proinflammatory monocytes and Alb(r=-0.2192,p=0.0022),C3(r=-0.4327,p=0.0432)and GFR(r=-0.6435,p<0.0001);positively with the level the expression of proinflammatory monocytes and BUN(r=0.3393,p=0.0089)and proeinuria(r=0.6263,p<0.0001),while there were no correlation with Ig G?Ig M and Ig A.The proinflammatory monocytes subpopulations detected to be secrete more inflammatory cytokines.Through the treatment,there was a marked drop in the expression of proinflammatory monocytes(p=0.004).Significance and Innovation: Compared with HC,the expression of proinflammatory monocytes on monocytes were higher in LN than in HC,and the proinflammatory monocytes secrete more inflammatory cytolines than the other groups.It was correlated with their expression the level of disease activity index.The results proved that proinflammatory monocytes might play a role in the pathogenesis of LN. |
