Objective:To study the changes of SOD activity and macrophage-secreted cytokines of Candida glabrata under the condition of macrophage engulfing Candida glabrata.Method:1.The Candida glabrata strains from different diseases were collected and identified by molecular biology,and a standard strain was added as a research strain.2.RAW264.1 macrophages were co-cultured with experimental strains.3.SOD activity assay kit was used to determine the superoxide dismutase activity of Candida glabrata phagocytosed by macrophages at different time points,and the changes of SOD values at different time points were observed.4.The ELISA kit was used to determine the macrophage cytokine secretion.5.The susceptibility testing of the strains was performed using an improved M27-A protocol recommended by the National Committee for Clinical Laboratory Standardization(NCCLS).6.Pretreatment of macrophages with an antifungal drug at a concentration lower than the MIC of each strain,co-culture with corresponding experimental strains,determination of SOD enzyme activity of Candida glabrata and macrophage IL-6,IL-8,TNF-a.7.Use SPSS software to analyze the effect of macrophage phagocytosis of superoxide dismutase and macrophage cytokine secretion after macrophage phagocytosis with antifungal drugs and without macrophages.Result:1.After phagocytosis of Candida glabrata by macrophages,its SOD activity values gradually increased with time and then decreased.2.Itraconazole and amphotericin B promoted the increase of SOD activity of macrophage in Candida glabrata compared with non-antifungal drug(Statistical analysis results were F=6.026,p<0.05;F=5.768,p<0.05);compared with no drug treatment,micafungin inhibited SOD activity of the strain(F=9.216,p<0.05).3.The secretion of IL-6,IL-8 and TNF-a by macrophages after phagocytosis of Candida glabrata was significantly higher than that of Candida glabrata without phagocytosis.4.There was a significant difference in the secretion of TNF-a from the macrophage between the micafungin group and the untreated group(t=6.541,P<0.05).After the phagocytosis of the strain,the secretion of TNF-a in the macrophage of the micafungin group was decreased,and there was no significant difference in other cytokines.The secretion of the three cytokines in the itraconazole group and the amphotericin B group was compared with that of the non-drug group.No statistical difference.Conclusions:After the phagocytosis of Candida glabrata by the macrophage,the SOD enzyme activity of the experimental strain increased with time,peaked at 6h,and then gradually decreased.2.The pretreatment of macrophages with itraconazole and amphotericin B promoted the increase of SOD activity of strains of Candida glabrata phagocytosed by macrophages,while the inhibitory effect of micafungin drug on the activity of SOD enzyme was inhibited.effect.3.Macrophages secrete IL-6,IL-8 and TNF-a over time after phagocytosis by Candida glabrata.4.Micafungin has the effect of down-regulating the secretion of TNF-a in macrophages under the condition of non-phagocytic Candida phagosa.It has no effect on the secretion of IL-6 and IL-8;itraconazole and amphotericin B are the other three The secretion of these cytokines had no effect.5.Micafungin reduced the secretion of IL-6,IL-8 and TNF-a by macrophages after phagocytosis of Candida glabrata.Itraconazole and amphotericin B attenuated Candida glabrata.The phagocytic cells secreted IL-6,IL-8 and TNF-a had no effect.
