Objective:This study is to explore the methylenetetrahydrofolate reductase(MTHFR)gene C677T and A1298C single nucleotide polymorphisms(SNP)mutation once diagnosed in the Calmette Affiliated Hospital of Kunming Medical University and their association with granulomatous lobular mastitis(GLM),for providing new clues for etiology and prevention of granulomatous lobular mastitis(GLM).Methods:61 female diagnosed with granulomatous lobular mastitis(GLM)and 60 healthy female subjects(control)were recruited in the Calmette Affiliated Hospital of Kunming Medical University between Sept,2016 and Dec,2017.Two groups age was no statistical difference.,ages of 17-48 years old(average 31.1).All subjects were selected with 2ml forearm venous blood for DNA extraction and MTHFR gene polymorphism detection.The polymorphisms of C677T and A1298C in MTHFR gene were detected by real-time fluorescence PCR using Taqman probe.The correlation between MTHFR gene C677T and A12968T polymorphism and the granulomatous lobular mastitis(GLM)was analyzed by case-control study.For x2 statistics test was applied,and P<0.05 was considered statistically significant.Results:Genotype CC,CT and TT are three genotypes of the C677T site of the MTHFR,and the allele T is the mutation.The prevalences of CC,CT,TT genetype of MTHFR gene C677T in GLM and the control group were 0.246,0.557,0.197 and 0.467,0.367,0.166,respectively.C/C genotype and C/T genotype have significant differences between the two groups(P=0.011,P=0.035),T/T genotype has no statistically significant difference between the two groups(P=0.668).Allele frequency of C,T in GLM group and control group were 0.525,0.475 and 0.658,0.342 respectively,and the differences between two groups was statistically significant(P=0.048).Genotype AA,AC and CC are the three genotypes of the MTHFR A1298C site,and the allele C is the mutated gene.The prevalences of AA,AC,CC genetype of MTHFR gene A1298C in GLM and the control group were 0.721,0.246,0.033 and 0.683,0.267,0.050,respectively.The frequencies of A/A,A/C and C/C were no statistically difference between the two groups(P = 0.648,P = 0.794,P =0.634).Allele frequency of A,C in GLM group and control group were 0.844,0.156 and 0.817,0.813,respectively,and there was no difference between the two groups(P=0.567).Conclusiolts:1.The SNP of the MTHFR gene C677T were associated with GLM.2.There was no correlation between the polymorphisms of the MTHFR gene A1298C site and the GLM.
