| Object:To investigate the effects of Yinchenwuling Powder on NAFLD and expression of Chemerin,CYP2E1 and NLRP3 in rats.Methods:60 male SD rats were divided into the normal group and the experimental group according to the random number table method.The experimental group was fed with high-fat diet for 24 weeks.After successful modeling,the rats were randomly divided into model group,simvastatin control group and high-dose Yinchenwuling Powder and low-dose Yinchenwuling Powder.The rats' body weight,liver's weight,liver index,liver serum biochemical indexes(ALT,AST,TC,TG,and LDL-c)of each group were measured after a six-week drug intervention;The pathological changes of liver tissue were observed after HE staining on the liver tissues;The protein expressions of rats' adipose factor Chemerin were tested by ELISA.The gene expressions of liver CYP2E1 and NLRP3 were tested by Real-time PCR.Result:1.Compared with the normal group,the body weight,liver wet weight,liver index and hepatic lipids in the model group were increased(P<0.01),and the Chemerin,CYP2E1,NLRP3 protein,and m RNA expression were increased(P<0.01),too.2.Compared with the model group,the body weight,the liver wet weight,the liver index and the liver blood lipid were decreased in the positive drug group and in the each dose group of the Yinchenwuling Powder(P <0.01),and the Chemerin,CYP2E1,NLRP3 protein,and m RNA expression were all decreased(P<0.01).3.Compared with the simvastatin positive group,the liver wet weight,the liver index and the liver blood lipid were decreased in the each dose group of the Yinchenwuling Powder(P<0.05,P<0.01,P>0.05),and the Chemerin,CYP2E1,NLRP3 protein,and m RNA expression were all decreased(P<0.01,P>0.05).Moreover,high-dose Yinchenwuling Powder group was better than that of low-dose Yinchenwuling Powder group.Conclusion:Yinchenwuling Powder can down-regulate the expression ofChemerin,CYP2E1 and NLRP3 protein,reduce liver lipid accumulation and improve liver function. |
