background and objectiveDysfunction of cognition,exercise,language and swallowing are common in stroke patients,greatly impairing patients' ability to take care of themselves and reducing the quality of life of patients,which is an issue that can not be neglected by individuals,families and the society.According to the research at home and abroad and the basis of previous research of our team,infrasound with low sound pressure can improve the functional status of rats with cerebral ischemia.Low-sound pressure infrasound through the mechanical resonance can dredge the tiny blood vessels,reduce the degree of cerebral infarction and promote the connection between neurons in vitro can promote the proliferation of stem cells,and because of its strong penetrating properties,even with skull barrier wrap,can still penetrate the skull to transmit mechanical resonance to intracranial tissue.Therefore,this experiment aims to establish the model of focal cerebral ischemia in rats experiment to observe the changes of cognitive and motor function after focal low-sound infrasound after focal cerebral infarction in rats,and by immunofluorescence labeling to observe the expression of Brdu and Nestin,a marker of proliferation of NSCs in brain tissue,and to explore the mechanism of protection of the nervous system by infrasound with low sound pressure,providing a new treatment for blood diseases.MethodThe right middle cerebral artery occlusion(MCAO)rat model was prepared and treated with infrasound 8TM infrasound as the treatment factor.Thirty-six Sprague-Dawley rats were randomly divided into three groups:Sham-operation group which without embolization(n = 12),operation group(n = 12)and infrasound group(n = 12).After 12 hours of successful modeling,the infrasound group started infrasound treatment for 1 hour per day,and the remaining two groups were placed under the action probe but no energy was output.The improved neurological deficit score of each group was observed on the 3rd,7th and 14th days after the intervention.Morris water maze training was performed on the 3rd to 6th day after the model was established.The escape latency of the rats in each group was recorded and on the last day the number of rats across the platform was recorded.Rats in each group were intraperitoneally injected with Brdu 32 hours before sacrifice at three time points,each time for 4 hours for 3 times.24 hours after the last injection,the brains were perfused and the sections were stained with immunofluorescence double staining to detect the BrdU positive cells and Nestin positive optical density(IOD)values.ResultCompared with the surgery group,the infrasound group at three time points of cognitive function and motor function were significantly improved(P<0.05).The expression of neural stem cells on the 3rd,7th and 14th day were respectively infrasound group>surgery group>sham operation group,the differences were statistically significant(P<0.05).With the increase of treatment days,the proliferation of neural stem cells in infrasound showed an increasing trend,with statistical significance(P<0.05).The proliferation of neural stem cells on the 7th day in the operation group increased compared with the 3rd day,the difference was statistically significant(P<0.05).The difference between day 14 and day 7 was not statistically significant(P>0.05).ConclusionInfrasound play a role of neuroprotection and repair by promoting the proliferation of NSCs in the brain tissue infarct,prolong the proliferation time,improve the cognitive and motor function.
