| Malaria,as the most important mosquito-borne infectious disease,has brought a heavy burden on the global health care.In addition,new mosquito-borne infectious diseases continue to emerge,Zika virus disease has caused several pandemics in the past ten years,and several imported cases of Zika virus infection have been reported in some coastal provinces in China.Descriptive analysis of malaria distributions in China were performed,malaria hotspots and local clusters were detected by spatial autocorrelation analysis,and scan statistics.Then,the incidence of malaria in Tanzania was fitted by bayesian spatial-temporal interaction model,and the effect and intensity of meteorological factors and social factors on the disease were discussed.Finally,the recombinant Zika virus envelope protein Ⅲ domain(rZED Ⅲ)was purified by prokaryotic expression in E.coli and purified by NTA to immunize mice.Specific monoclonal antibodies against zika virus were prepared and their specificity and immunological characteristics were studied.In this study,we found that the prevalence and intensity of malaria in China were significantly decreased,and gradually changed to imported cases after 2010;the aggregation of malaria from the three provinces of hainan,anhui and yunnan gradually reduced to yunnan border areas,the duration of clustering period was significantly shortened,and the correlation between meteorological factors in China and the incidence of malaria dropped sharply.In the mainland of Tanzania,the malaria prevalence intensity is in a downward trend as a whole,and is mainly concentrated in the eastern low-altitude areas.The type I bayesian spatio-temporal interaction effect model shows the best fit(DIC=44136),suggesting that the malaria incidence is not only susceptible to meteorological factors such as temperature(OR=1.034),rainfall(OR=1.003),wind speed(OR=0.9),and relative humidity(OR=0.998),but also affected by the socioeconomic factors as log population density(OR=0.616)and the confirmed diagnosis rate(OR=0.997).Up to now,three hybridoma cells capable of secreting specific monoclonal antibodies against zika virus have been screened in this study.IgG subtype has been identified as IgG1.Western Blot experiments and immunofluorescence experiments have found that mAb10b4 has the strongest binding ability and can bind to rZED Ⅲ and virus particles,which can provide support for the diagnosis and treatment of zika virus.In summary,imported malaria has become a major challenge in China,the association between malaria incidence and climate factors decreased after 2010,strengthen the diagnosis and treatment for Farmer&Worker from high risk areas should be more apporriate.In addition,malaria is still affected by the natural environment in Tanzania.The mosquito vector prevention and serology monitoring in areas with low temperature and rainfall,low temperature and high relative humidity in eastern low altitude regions should be improved.Besides,specific monoclonal antibody against zika virus envelope protein was preliminarily prepared and its immunological characteristics were identified,and mAb 10b is expected to become a candidate for diagnosis and treatment of zika virus. |
