Objective: To investigate the effect of PRMT2 gene on the apoptosis of human umbilical vein endothelial cells(HUVECs)and to explore its possible mechanism.Methods: In this study,HUVECs were used as experimental subjects.Firstly,high and low expressed plasmids of PRMT2 were transiently transfected respectively.Fluorescence inverted microscope was used to observe the expression of GFP green fluorescent protein in transfected cells.The expression of PRMT2 in cells was detected by PCR and Western blot.Then,the effect of PRMT2 on HUVECs and cell viability was observed by MTT assay.Moreover,the effect of PRMT2 on the migration rate of HUVECs was measured by wound healing assay.At last,Western blot was used to detect the expression of Bcl-2,Bax and caspase-3 in HUVECs induced by PRMT2.Results: 1)The expression of PRMT2-mi RNA and PRMT2 protein in HUVECs transfected with PRMT2 high expression plasmid was significantly increased.The expression of PRMT2-mi RNA and PRMT2 protein in HUVECs transfected with PRMT2 low expression plasmid was significantly reduced.2)High expression of PRMT2 significantly inhibited the viability and migration of HUVECs,up-regulated the expression of Bax and caspase-3,and down-regulated the expression of Bcl-2.3)The low expression of PRMT2 obviously increased the viability of HUVECs,and significantly increased the migration of HUVECs,as well as down-regulated the expression of Bax,caspase-3 and up-regulated the expression of Bcl-2.Conclusion: PRMT2 can inhibit the proliferation and migration of vascular endothelial cells,which may be related to the process of apoptosis.
