Objectives:This study was set to illustrate the absorption,metabolism,distribution and metabolism of the main bioactive components,namely Balanophorin B,gallic acid,stearic acid,and metabolism of stearic acid after oral administration of Balanophora Simoensis S.Y.Chang et Tam ethanol extract in vivo in rats.Methods:Rats were orally administered with ethanol extract of Balanophora Simoensis S.Y.Chang et Tam and stearic acid respectively.Blood samples and organ samples were collected at different time points,bile samples were taken at intervals of 1 h for 12 h;urine and feces was sampled at intervals of 24 h for 96 h.The crude extracts and Balanophorin B,gallic acid and stearic acid were detected after oral administration in vivo in rats.All samples were analyzed by UHPLC-MS/MS after pretreatment.Results:After rats were orally administered with ethanol extract of Balanophora Simoensis S.Y.Chang et Tam,Balanophorin B,gallic acid and stearic acid were detected in blood samples;the substrates were detected in kidney,liver and bile;gallic acid Biotransformated in vivo,its phase I metabolite is 4-hydroxybenzaldehyde.It was not deduced that metabolic transformation occurred to the Balanophorin B and stearic acid in the samples.Balanophorin B,gallic acid and its metabolites 4-hydroxybenzaldehyde,stearic acid were identified in urine and feces samples.Stearic acid was detected in the plasma,kidney,liver,heart,brain,bile,urine and feces after oral administration of stearic acid.Conclusions:Balanophorin B,gallic acid and stearic acid were detected in the crude of Balanophora Simoensis S.Y.Chang et Tam.Balanophorin B,gallic acid and stearic acid had also been identified in plasma,kidney,liver,bile,urine and feces samples after oral administration by UHPLC-MS/MS.The retention time of gallic acid is 11.792±0.5min,it was metabolized,its phase I metabolites was 4-hydroxybenzaldehyde with the retention time of 2.194 min±0.5 min.Balanophorin B and stearic acid were not found metabolic transformation in the samples.Stearic acid was detected in the plasma,kidney,liver,heart,brain,bile,urine and feces samples after oral administration of the standard stearic acid,and it was supposed to penetrate the blood-brain barrier.
