Objective:With the development of our nation's economy and the progress of the society,people's living standards continue to improve.At the same time,changes in the diet habits have brought about aging problem.The rate of morbidity and mortality of colorectal cancer also rises,which seriously affects the health of the human body.Studies show that colorectal malignant tumor in the human body changes with different periods,in relation to inflammatory hyperplasia,the change of adenoma canceration,carcinogenesis and metastasis,etc.Long-term experimental observation has found that tumor cells have the ability of infinite proliferation and independent growth.Hence it is of great significance for cell proliferation and independent separation biomarkers to guide and evaluate the tumor occurrence and deterioration.Minichromosome maintenance proteins 2(MCM2)only exists in eukaryotic nuclei,eukaryotic cells which is the necessary complex components before DNA replication.MCM2 is expressed in active split proliferation cells.It is verified that the expression increases significantly in mutagenic cells and the abnormally proliferative developmental cell.However,no expression or low expression is found in the differentiated cells and static period cell.This experiment intends to explore the different expressions of cell proliferation marker MCM2 in colorectal cancer,colorectal adenomas and normal mucosa tissues,and its relationship with different clinical pathological features.What's more,the present study aims at proving the early warning role of MCM2 in colorectal adenomas process of malignant transformation.Methods:1.Materials: Immunohistochemistry method is adopted to detect the expression parts of MCM2 in colorectal cancer,colorectal adenomas and normal intestinal mucosa tissue.The samples come from 16 cases of normal intestinal mucosa tissue,16 cases of colorectal cancer and 80 cases of Colorectal cancer tissue.The expression differences of MCM2 are analyzed.The 80 cases of malignant tumor tissue are divided according to patients' gender,age,tumor diameter,differentiation degree,infiltration depth and TNM staging.And an analysis is made on the relationship between the protein and clinical pathological factors.At the same time,the expression of Topo-??EGFR and P53 protein in malignant tumor is collected.Then a comparison is made between the expression level of MCM2 protein and its possible correlation between the protein expression.2.Observing parameter: The pathological changes and histological differentiation of the tissues are observed by HE staining.The location and expression of MCM2 in different tissues are detected by immunohistochemical method.3.Data and analysis: Data is analyzed by SPSS19.0.Measurement data are analyzed by variance analysis or t test.Chi-square test or rank sum test were used to count data and rank data.P<0.05 has statistical significance.Results:1.The level of MCM2 expression in normal colorectal mucosa group is lower than that in colorectal adenoma group,and lower than that in colorectal cancer group.The difference is statistically significant.2.There is no significant correlation between the expression level of 2.MCM2 and the age,sex and tumor diameter of colorectal cancer patients.There is no statistically significant difference(P > 0.05),but it is related to the pathological grade,depth of infiltration,lymph node metastasis and TNM stage of the tumor(P< 0.05).3.The expression of MCM2 is correlated with the expression of TopoII protein,EGFR protein and P53 protein,and the difference is statistically significant.The results show that with the enhanced expression of TopoII protein,the positive rate of MCM2 expression decreases gradually,and the expression of the two is negatively correlated,and the difference is statistically significant(P=0.039<0.05).As the expression of EGFR protein increases,the positive rate of MCM2 expression increases gradually,and the expression of the two is positively correlated,and the difference is statistically significant(P=0.017<0.05).With the increase of P53 protein expression,the positive expression rate of MCM2 decreases gradually,and the expression of two is negatively correlated,and the difference is statistically significant(P=0.001<0.05).Conclusion:1.Due to the differences in the expression of MCM2 in normal mucosa,colorectal adenoma and colorectal adenocarcinoma,it is suggested that MCM2 has a correlation with the malignant transformation of colorectal adenoma and the occurrence of colorectal cancer.2.The expression level of MCM2 and colorectal cancer pathology classification,infiltration depth,whether associated with lymph node metastasis,tumor were significantly correlated with TNM stage,show MCM2 can be used to prompt the malignant degree and the prognosis of patients with colorectal cancer.3.MCM2 and Topo in colorectal cancer group ?,EGFR and P53 protein in the correlation,through the analysis of the joint detection and more protein,such as its expression level will help further prompt the occurrence and development of colorectal cancer,transfer
