Objective: To compare the expression of serum ANGPTL8 before and after fat meal in population with different lipid metabolism, and to explore the relationship between ANGPTL8 and non-alcoholic fatty liver. Methods: This study was conducted in Endocrinology Department of Hebei Province General Hospital from May to December,2016.Eventually,a total of 113 individuals were enrolled in the study. All subjects provided written informed consent and took an oral fat meal test of 1500 kcal load.Venous blood samples of fasting and postprandial at 4-hour were taken for glucose and lipid metabolism index.Serum ANGPTL8 was measured by enzyme-linked immunosorbent assay before and after fat meal in all volunteers.Non-alcoholic fatty liver diagnosis based on ultrasound according to guidelines for diagnosis and treatment of nonalcoholic fatty liver disease in2010.The subjects were divided into 3 groups: normal fat tolerance(NFT),impaired fat tolerance(IFT),hypertriglyceridemia(HTG).SPSS21.0 software was used to analyze the relationship between ANGPTL8 and nonalcoholic fatty liver. Results: The level of fasting ANGPTL8 in NFT group was higher than IFT and HTG group, with statistical difference(P=0.002,P<0.001).The level of fasting serum ANGPTL8 was negatively correlated with fasting total cholesterol,triglyceride,Low-density lipoprotein cholesterol, apolipoprotein B,fasting blood glucose, fasting insulin and homeostasis model assessment of insulin resistance(P=0.001?P<0.001?P=0.001?P<0.001?P<0.001?P=0.038?P=0.006,r=-0.303?r=-0.503?r=-0.310?r=-0.323?r=-0.348?r=-0.195?r=-0.259,respectively).The incidence of non-alcoholic fatty liver gradually increased among the three groups, and the difference wasstatistically significant(P=0.003,P < 0.001,P<0.001,respectively).According to serum ANGPTL8 quartiles, volunteers were divided into four groups. From Q1 group to Q4 group, the incidence of non-alcoholic fatty liver was 51.72%,42.86%,35.71%,and 10.71%,respectively?Compared with Q4 group, the incidence of fatty liver in Q1?Q2 ?Q3 group was statistically significant(P=0.003,P=0.018,P=0.027,respectively).Conclusion: The level of serum ANGPTL8 is closely related to the occurrence of non-alcoholic fatty liver, and as the level of ANGPTL8 decreases, the incidence of non-alcoholic fatty liver increases.
