Objective:Postn has a key role in the pathological process of various fibrotic diseases and the pro-fibrogenic factor TGF-?1 signaling pathway.However,the function of Postn in IUAs was unclear.To investigate the roles of Postn,?-SMA and Collagen I in IUAs and the relationship with the severity of IUAs,by means of detecting the mRNA and protein expression of Postn,?-SMA and Collagen ? in endometrium tissues from patients with different degrees of IUAs,and the protein production of them induced by TGF-?1 in primary ESCs.Method: A total of sixty human endometrial biopsy samples(mild IUAs:20,moderate IUAs:20,severe IUAs:20)were collected with confirmed IUAs by hysteroscope(experimental groups),plus 20 patients without IUAs(control group).The mRNA and protein expression of Postn,?-SMA and Collagen ? in different severity IUAs patients respectively were detected by qRT-PCR,Western blotting and IHC staining analysis.As well,we detected the protein levels of Postn,?-SMA and Collagen I in primary ESCs treated with TGF-?1 by Western blotting.Results QRT-PCR,Western blotting and IHC staining analysis showed that the mRNA and protein expression of Postn were positively correlated with the expression of ?-SMA and Collagen I and the severity of intrauterine adhesions.We also found that the levels of Postn,?-SMA and Collagen I in the experimental groups were higher than the control group,especially the moderate and severe IUAs groups(P < 0.01).Western blotting analysis revealed that there was a time-and dose-dependent manner between TGF-?1 stimulation and the protein levels of Postn,?-SMA and Collagen ? in cell experiments.Conclusion: The results confirmed that the up-regulation of Postn,?-SMA and Collagen I induced by TGF-?1 participated in the fibrotic process of IUAs,and was related to the severity of IUAs.We speculated that the pathomechanism of IUAs in response to endometrial injury may be related to myofibrosis of ESCs,which could be promoted by continuous high expression of Postn upon TGF-?1 stimulation.
