Objective: Genome-wide screened differentially expressed long non-coding RNA(lncRNA)in esophageal squamous cell carcinoma(ESCC)of Han nationality in Xinjiang,and investigate the transcriptome characteristics and biological functions of related lncRNAs in ESCC.Methods: Six endoscopic specimens of esophageal squamous cell carcinoma and adjacent normal tissues from Xinjiang Han people were collected.The total RNA was extracted and its quality was tested.After reverse transcriptase synthesis of c DNA,fluorescence hybridization was performed on a human long-chain non-coding RNA(lncRNA)V4.0 Agilent chip containing 40,916 lncRNAs detection probes and 34,423 m RNA detection probes,and then processed using a chip scanner.The chip scanning identification,acquisition of hybrid pictures,hybridization images using Agilent(v10.7)biological characteristics extraction software to identify,analyze and extract the original data,the original data were normalized and the difference analysis,and finally obtained differentially expressed lncRNAs.The lncRNAs-m RNAs co-expression network was constructed,target gene prediction was performed,and further studies were performed using GO(Gene Ontology,Gene Ontology)and Pathway analysis.Results: In esophageal carcinoma and its adjacent normal tissues,680 lncRNAs were identified(more than2 changes and P<0.05),of which 161 were up-regulated and 519 were down-regulated;1742 m RNAs were differentially expressed,of which 653 were upregulated of 819 m RNAs were down-regulated.15 differentially expressed lncRNAs had target genes,of which 11 lncRNA regulated the target genes by cis-regulation.The target genes were ANKRD20A2,lnc-GLIPR1-3,DLEU1,C5orf66-AS1,GALNT12,IGFL1,et al.One lncRNA has a trans-transtransregulatory target gene,the target gene is MALL.And three lncRNA can regulate the target genes by cis-and trans-transgene,and the target genes are ARSGN,MPZL3 and ATG9B.The GO enrichment analysis found that 62 items were enriched,and among the top 30 enriched items,there were 4 molecular functions,14 cellular components,and 12 biological processes.Ranked first and second are the cellular components of the Golgi membrane and the autophagic vacuole membrane,and the third is the molecular functional polypeptide N-acetylgalactosamine transferase.The KEGG,Reactome,and Bio Cyc pathway assays demonstrated that target genes for lncRNAs are involved in11 pathways that may be associated with esophageal squamous cell carcinoma,including post-translational protein modifications,mucin O-glycosyl biosynthesis,activation of arylsulfatase,and sheath sugar and lipid metabolism pathways and other pathways.Conclusion: There are a large number of differentially expressed lncRNAs in esophageal squamous cell carcinoma of Han nationality in Xinjiang.These differentially expressed lncRNAs may play an important role in the development of esophageal carcinoma through some signaling pathways,the interaction of lncRNA and m RNA may reveals a complex regulatory network of esophageal carcinoma.It is of great significance to find more effective new targets for diagnosis and treatment of esophageal squamous cell carcinoma.
