Promoting the regeneration of damaged nerves has always been a difficult and hot topic in today's research.Chitosan and alginate as natural polysaccharides have good biocompatibility and are widely used in the field of nerve repair.However,these two materials alone have certain limitations and functional self-assembling peptide nanofibers hydrogels(F-SAP)can promote nerve growth and have potential applications in the field of nerve regeneration.Basing on previous studies,we prepared chitosan/calcium alginate/F-SAP hydrogel and studied its application in nerve neural regeneration.A bio-crosslinker genipin cross-linked chitosan and F-SAP to obtain a chitosan/F-SAP scaffold;FSAP/sodium alginate solution reacted with calcium chloride solution,after the hydrodynamic stretching,the parallel active hydrogel microfibers was obtained;the chitosan/calcium alginate/F-SAP hydrogel microfibers was obtained through polyelectrolyte composite reaction.The physicochemical properties were characterized by SEM and polarized light microscopy,etc.;The application of the nanocomposite scaffold in nerve regeneration was studied by cytotoxicity,the interaction between the matrixt and the neural stem cells.The results of the study are as follows:(1)The chitosan/F-SAP scaffold material has a porous structure,and the optimum parameters for obtaining the optimum volume ratio of the two materials are 1 ? 1.The results of cell biology safety evaluation showed that chitosan/F-SAP scaffold had good biocompatibility.(2)A parallel hydrogel microfibers were prepared by optimizing the parameters.Concentrations of sodium alginate and calcium chloride and their respective injection rates have an effect on the hydrogel microfibres.The diameter of the hydrogel microfibres could be controlled between 20~40 ?m,and the moisture content was above 90%.The prepared samples were highly regular and parallel.(3)The biocompatibility of chitosan/calcium alginate/F-SAP hydrogel microfiber had been improved.The results of cell immunostaining showed that scaffold materials could guide the growth of neurons.The nerve scaffold material prepared in this study has good biocompatibility,can support the growth and differentiation of neural stem cells and can guide the directional growth of axons.Thus it has a good application prospect in the field of nerve repair.
