Objective:Observing the effect on mark metabolites of long-chain fatty acid of electroacupuncture treatment of “HT7”and “SP6”on patients with insomnia;Exploring the mechanism of electroacupuncture treatment of “HT7” and “SP6”of insomnia.Methods:1.Divideding 20 patients with insomnia into insomnia group and HT7-SP6 group randomly,and 10 healthy people as healthy group.HT7-SP6 group was treated with electroacupuncture treatment of “HT7”and “SP6”,this treatment was continued for 2 courses,5 days for one course and once a day.2.Evaluating the quality of sleep by PSQI and drowsiness the next day by ESS.Observing the effect on long-chain fatty acid of electroacupuncture treatment of “HT7”and “SP6”on patients with insomnia by using LC-MS/MS metabolomics technology to detecte serum metabolites.Observing the regulatory mechanism of electroacupuncture treatment of “HT7”and “SP6”on patients by using LC-MS/MS metabonomics.Results:1.After 2 courses of electroacupuncture treatment of “HT7”and “SP6”,the total score of PSQI in HT7-SP6 group was decreased from 11.40±1.65 to 5.60±2.41,the difference has statistical significant;The total score of ESS in HT7-SP6 group was decreased from 7.60±4.77 to 6.30±5.70,but has no statistical significant.2.Compared with healthy people,the relative concentrations of(R)-2-Hydroxyhexadecanoic acid,(R)-3-Hydroxyhexadecanoic acid,3-Hydroxyhexadecanoic acid,(±)-3-[(2-Methyl-3-furyl)thio]-butanone,(R)-3-hydroxymyristic acid,2-Hydroxymyristic acid,3-hydroxytetradecanoic acid,3-(L-Menthoxy)-2-methylpropane-1,2-diol,Ethyl 3-hydroxydodecanoate,10,20-dihydroxyeicosanoic acid in the serum of patients with insomnia were increase.Among them,(R)-2-hydroxyhexadecanoic acid,(R)-3-hydroxyhexadecanoic acid,3-hydroxyhexadecanoic acid,(R)-3-hydroxymyristic acid,2-Hydroxymyristic acid,3-hydroxytetradecanoic acid and 10,20-dihydroxyeicosanoic acid are long-chain fatty acids.3.After 2 courses of electroacupuncture treatment,the relative concentrations of(R)-2-hydroxyhexadecanoic acid,(R)-3-hydroxyhexadecanoic acid,3-hydroxyhexadecanoic acid,(R)-3-hydroxymyristic acid,2-Hydroxymyristic acid,3-hydroxytetradecanoic acid and 10,20-dihydroxyeicosanoic acid were all down-regulated in serum of patients with insomnia;Among them,the relative concentrations of(R)-2-hydroxyhexadecanoic acid,(R)-3-hydroxyhexadecanoic acid,3-hydroxyhexadecanoic acid and 10,20-hydroxyeicosanoic acid were no difference with healthy group.Conclusions:1.Electroacupuncture treatment of “HT7”and “SP6” can improve the quality of sleep in patients with insomnia.2.There were significantly increased of(R)-2-hydroxyhexadecanoic acid,(R)-3-hydroxyhexadecanoic acid,3-hydroxyhexadecanoic acid,(R)-3-hydroxymyristic acid,2-Hydroxymyristic acid,3-hydroxytetradecanoic acid and 10,20-dihydroxyeicosanoic acid in serum of patients with insomnia.And they are potential long-chain fatty acid metabolism markers of insomnia.3.Electroacupuncture treatment of “HT7”and “SP6”can adjust the potential long-chain fatty acid metabolism markers in patients with insomnia,especially for(R)-2-hydroxyhexadecanoic acid,(R)-3-hydroxyhexadecanoic acid,3-hydroxylhexadecanoic acid and 10,20-hydroxyeicosanic acid,this treatment can restore their relative concentrations to normal levels.
