Comparative Study On Icariin And Bu-Shen-Gu-Chi-Wan For Alveolar Reconstruction In Chronic Periodontitis Of Rats

Abstract/Summary:
Objective: Observation the effects of icariin(ICA)and Bu-Shen-Gu-Chi-Wanon the alveolar bone absorption of chronic periodontitis in rats,and further exploration the effect of ICA on chronic periodontitis and its possible mechanism.Methods: Sixty healthy male Sprague Dawley(SD)rats aged 7 weeks were selected.A total of 51 rats were randomly selected.The second maxillary right maxilla was used as the experimental tooth,and the ligament + high glucose diet was used to establish the model of periodontitis.The remaining nine untreated and given a normal diet,the normal group.After 2 months,3 rabbits in periodontitis model group and 3 normal rabbits were sacrificed.The oral cavity examination,Micro-CT and routine sliced periodontitis models were successfully established.The remaining 48 SD rats of periodontitis model group were randomly divided into four groups which was fed a normal diet with12 rats.ICA low dose group :ICA 20 mg /(kg · d)ig,ICA high Dose group: 40 mg /(kg · d)ig,Bu-Shen-Gu-Chi-Wanl group: Bu-Shen-Gu-Chi-Wan 0.8g /(kg · d)ig,periodontitis group:double distilled water 10 ml /(kg · d)ig.One week after the basic treatment,normal group was given a normal diet with double distilled water 10 ml /(kg · d)in gavage.In 1st month and 2th month in gavage,the normal group were sacrificed with 3 rats and the remaining groups were sacrificed 6.Carotid artery blood was taken to detect serum bone turnover markers: serum osteocalcin(OCN)and alkaline phosphatase(ALP).The alveolar bone in the right experimental area was scanned by micro-computed tomography(CT)to observe the new bone mass and attachment loss of experimental alveolar bone in each group.Next,each experimental tooth was divided into two parts in the near and far directions.After the scanning,the histological sections(HE staining)and the histological sections(toluidine blue staining)were made.The alveolar bone,cementum and periodontal ligament were observed.Results: 1、Compared with the normal group,there were significant decreases in the levels of ALP and increased serum OCN of the periodontitis group(p<0.05).ICA-low、ICA-high dose groups,the Bu-Shen-Gu-Chi-Wan treatment group had significant decreases in the levels of serum OCN and increased serum ALP than the periodontitis group(p<0.05).2、Quantitative analysis of bone parameters suggested,In the periodontitis group,the BV/TV,Tb.N,and Tb.Th values in the ROI were significantly reduced,and the Tb.Sp value was significantly increased(p<0.05),while the three drug treatment group increased BV/TV、Tb.N、Tb.Th values,and decreased Tb.Sp value(p<0.05).In 2st month,the Tb.N value of the alveolar bone in drug treatment groups were significantly higher than those in 1st month and the Tb.Sp value were lower(p<0.05).Morphological analysis shownthe alveolar bone resorption value in ICA-low、 ICA-high dosegroups and the Bu-Shen-Gu-Chi-Wan treatment group was significantly lower than that in the periodontitis group.3、According to HE stain and toluidine blue stain,the periodontitis group showed obvious periodontal inflammationand the periodontal supporting tissues were severely damaged.After treatment with ICA and Bu-Shen-Gu-Chi-Wan,periodontal inflammation was reduced,and periodontal repair and regeneration were active.Conclusion:1、ICA can improve the alveolar bone resorption of experimental chronicperiodontitis.Both ICA and Bu-Shen-Gu-Chi-Wan can repair periodontal tissue defects and promote periodontal regeneration,and there is no significant difference in curative effect of the two.2、ICA may relieve tissue destruction and promote alveolar bone remodeling by regulating periodontal bone metabolism and othermechanisms,and may has a certain role in adjuvant treatment of periodontitis.
