Objective:To investigate the expression of LncRNA DANCR in placenta of women with severe preeclampsia(SPE)and its effect on the trophoblast biological behavior.Methods:Real-time quantitative PCR was used to detect the expression of Lnc RNA DANCR in placenta of 20 pregnant women with severe preeclampsia from 36 weeks to 39 weeks and 20 normal pregnant women with 37 weeks to 39 weeks.By culturing HTR-8 / SVNEO trophoblast cells and using gene interference technique,the changes of trophoblasts in the biological behavior after the decrease of DANCR expression were detected.Results:1.In severe preeclampsia,compared with normal pregnant women,the expression of DANCR in placenta was significant decreased(N=1,PE=0.4639±0.27576,P<0.05).2.After si RNA-transfected cells were transfected with si-DANCR,the expression of DANCR and NC group in si-DANCR transfected cells was significantly down-regulated compared with the control group.3.According to the cell growth curve drawn by MTT method,compared with the control group,HTR-8/SVNEO cells which DANCR downregulated began to show a decrease in proliferative capacity 48 hours after transfection.4.After 48 h transfection,cells were scratched test cell migration ability and found migration ability of HTR-8/SVNEO cells which DANCR down-regulated is weakened.5.After 48 h transfection,the number of invasive cells in si-DANCR transfected group was significantly less than that in control group,and the invasion index was significantly lower than that in control group(P <0.001)at the same time.Conclusion: The expression of Lnc RNA DANCR in placental tissue of patients with severe preeclampsia significantly decreased,which may be involved in the pathogenesis of preeclampsia by regulating the proliferation and infiltration of trophoblasts.
